Objective: This paper intends to carry out in-depth study on the traits,identification,inspection,content determination and characterization of the leaves of Mahonia bealei(Fort.)Carr,and make a comprehensive evaluation of the quality of herbs and establish a scientific,stable and controllable quality standard.Methods : The use of eye,hand touch,smell,taste and other sensory methods of untreated medicinal herbs to observe and judge.The micro-structure of herbs can be identified by high power microscope.The basic chemical constituents of berberine,palmatine and jatrorrhizine were used as the reference substance,and the best thin layer chromatographic conditions were explored to establish a simple and rapid thin layer chromatography method.HPLC method was used to determine the content of the leaves of Mahonia bealei(Fort.)Carr in berberine,palmatine and jatrorrhizine as the reference substance.The results were statistically calculated and the content limit was determined.According to the 2015 edition of "Chinese Pharmacopoeia" contained methods of determination of water,total ash and extract content,and provides its limits.Based on the chromatographic conditions of the content determination,the conditions were further optimized.HPLC method was used to determine the leaves of Mahonia beale(Fort.)Carr of different batches.The chromatographic peaks were processed.By calculating the relative peak area and relative retention time and similarity,Create a map of the leaves of Mahonia bealei(Fort.)Carr.Results: The collection of ten batches of Mahonia bealei Leaves is divided by color,shape,gas,taste four aspects of trait description,can objectively reflect the quality characteristics of Pieces.The characteristics of upper and lower epidermal cells,sponge cells,stomata and fiber were observed under microscope,which was of great significance for the rapid identification of Pieces.The limits of 13%,5%,and 10% of the water content,total ash content and extract content of the 10 batches of Pieces are complementary to the quality of medical materials.Thin layer identification test,the use of ultrasound method to extract the chemical composition of the sample,easy to operate and the test solution for the spot Rf value is moderate,clear spots,the reference substance and the test sample spot position and color consistent,to confirm the berberine,palmatine and jatrorrhizine are the chemical constituents of the leaves of Mahonia bealei(Fort.)Carr.The content of berberine,palmatine and jatrorrhizine in Mahonia bealei Leaves were determined by HPLC at 346 nm.The chromatographic peaks of the three components reached the baseline separation,and the injection concentration was linear with the peak area.The average recoveries were 99.17%(RSD = 0.35%,n = 6),98.97%(RSD = 0.65%,n = 6)and 99.36%(RSD = 0.44%,n = 6),respectively.Based on the analysis of the relative retention time and relative peak area of each chromatographic peak of 10 batches of the Mahonia bealei Leaves,the 10 common peaks were analyzed by using the software of "Chromatographic Fingerprint Similarity Evaluation System A" in 2004.The similarity of samples G1 and G7 is about 0.80,and the similarity of the remaining 8 batches of medicine is above 0.95,which indicates that although different origin and growth environment have influence on the quality of medicinal materials,it can still be used to identify the medicinal materials through the characteristic map.The identification of Mahonia bealei Leaves is specific.Conclusions: Through the study of the leaves of Mahonia bealei(Fort.)Carr,hoping to make up for the lack of detection methods,to provide a feasible reference for the control of Pieces quality,the market circulation of Mahonia bealei Leaves for unified management,the establishment of multi-indicators,quantifiable evaluation criteria,And for the new version of " Chinese medicine Pieces Code of Anhui Province" to provide quality standards(draft)and instructions.
