Obiective: To understand the situation,distribution,infection and genetic evolution of viruses in ticks from Yunna Province,China.The results provide scientific guidance for prevention and comtrol of the related diseases.Methods: Firstly,the high-throughput sequencing method was used to obtain the virus coenosis of the tick samples,and the specific primers were designed according to the nucleotide sequence of the virus.RT-PCR method was used to detect the infection status of the tick-borne virus and the positive samples were used for virus isolation.The viral genome was amplified in some significant virus samples and the virus genetic evolution was analyzed by bioinformatics software.The virus nucleocapsid protein was expressed by the prokaryotic cells and developed the antibody angaist the expressed protein by immune rabbit methods.Results: The total number of nucleicacid reads obtained by high-throughput sequencing was 12596,and the six strains of viruses were obtained.One strain of virus was named Yunnan Tick Virus2(YNTV2)with Wuhan Tick virus 1(WHTV1)homology was 96%,which belong to Unclassified Dimarhabovirus 1.The infection rate of YNTV2: was Lushui County 45.24%(38/84),Binchuan County 16.13%(5/31),Luxi County 9.09%(4/44)and Eryuan County 5.26%(1/19)respectively in tick samples in four countries,Yunnan province.YNTV2 isolation was unsuccessful.The nucleocapsid protein of YNTV2 was expressed and purified by prokaryotic cells,and the antibody against the expressed protein was prepared.Conclusion: The virus coenosis is mainly Bunyaviridae,Rhabdoviridae and other unclassified viruses in 4 counties,Yunnan Province.The results showed thegenetic diversity of the virus in the investigated areas.Lushui County has the highest infection rate of YNTV2 in four survey counties in Yunnan.Purified YNTV2 nucleocapsid protein can be used for serological epidemiological investigations.
