Objective: to detect the expressions of CD134(OX40,TNFRSF4),CD134 ligand(CD134L,CD252,TNFS4)in pancreatic cancer tissues and adjacent normal pancreatic tissues and analyze the relationship between the expressions and clinicopathological features of pancreatic cancer and the prognosis of patients and explore the effect of CD134 and CD134 L on the microenvironment of pancreatic cancer immunity.Methods: 1.paraffin specimens of 42 cases in department of general surgery of yijishan Hospital,affiliated to Wannan Medical College were diagnosed as pancreatic cancer according to the gold standard of postoperative pathology were collected and the corresponding adjacent normal pancreatic tissue specimens were also collected from May 2012 to October 2015.2.the paraffin specimens of pancreatic cancer,were detected by immunohistochemical staining at department of general surgery of yijishan Hospital,affiliated to Wannan Medical College(En Vision two step method).The expressions of CD134 and CD134 L in adjacent normal pancreatic tissues were detected and its clinical and pathological characteristic(tumor size,There is no nerve area,vascular invasion,lymph node metastasis,differentiation degree,clinical stage)were analyzed and the relationship between the expressions of CD134 and CD134 L and clinical and pathological characteristic.3.collect 42 patients with pancreatic cancer clinical data(gender,age,there is no history of cancer,chemotherapy has no systemic disease),and follow-up(outpatient and telephone return visit)their survival time(survival time of patients after standard: from the first day until the date of death or last follow-up date of October 2016),a retrospective analysis of CD134,the relationship between CD134 L expression and clinicopathological parameters of patients with pancreatic cancer and prognosis.Results:1.CD134 and CD134 L were detected in pancreatic cancer tissues at different levels,and in adjacent normal pancreatic tissues were not expressed,there was significant difference between two groups(P<0.05).2.CD134 and CD134 L expression in pancreatic cancer cells,the positive expression of CD134 localized in the cytoplasm,the positive expression of CD134 L in the cytoplasm and membrane.;42 pancreatic cancer tissues,22 CD134 positive expression,20 negative expression,the positive expression rate of 52.4%;19 CD134 L positive,23 negative expression,the positive expression rate of 45.2%,the positive expression rate of CD134 was higher than that of CD134 L.3.CD134 molecules in pancreatic cancer tissue expression and tumor size,lymph node metastasis,clinical stage(ACJJ stage)was significantly related(P<0.05),the mass is bigger,the positive expression of CD134 in cancer tissues was higher;lymph node metastasis,CD134 in cancer in the positive expression rate is high;the greater the ACJJ staging,the positive expression rate is also higher,while age,gender,tumor growth site,degree of differentiation,there is no nerve vascular invasion and other factors with no significant correlation(P>0.05).4.42 cases of patients with median survival of 13 months,CD134 positive group median survival of approximately 9 months,the negative group median period of about 21 months,draw Kaplan-Meier survival curves between the two survival analysis,using the method of Log-Rank analysis showed that: the expression of CD134 positive group survival than negative expression group and short survival period(P<0.05);Cox regression multivariate survival analysis showed that clinical stage(P=0.003),vascular invasion(P=0.002)were independent prognostic factors affecting the survival of pancreatic cancer patients,and CD134 Molecular,age,sex,tumor size,tissue differentiation,perineural invasion,and lymph node metastasis were not independent prognostic factors(P>0.05)for survival in patients with pancreatic cancer.Conclusion: 1,CD134,the high expression of CD134 L in pancreatic cancer tissues,the body of CD134,CD134 L molecules are involved in the formation of.2 in pancreatic cancer,the expression of CD134 positive patients survival than patients with negative expression of short survival,and the positive expression rate and clinical stage,tumor size,lymph node metastasis,indicating the special significance of.3 evaluation of CD134 expression in pancreatic cancer malignancy and prognosis,lymph node metastasis,vascular invasion are independent prognostic factors affecting the survival of pancreatic cancer patients.
