Objective:Immune cells infiltrated in tumors are related to tumor invasion,progression,and metastasis,but the role of immune cells in pancreatic cancer is not clear.Therefore,we analyze the correlation between immune infiltration characteristics and clinical characteristics and prognosis of pancreatic cancer.Materials and Methods:Download gene transcription profile data and clinical data of pancreatic cancer samples from TCGA database.The duplicated gene expression data were averaged and retained,and the remaining duplicates and genes with 0 expression levels were deleted.After correction,gene transcription profile data of 182 pancreatic cancer samples were obtained,of which 178 were tumor tissues and 4 were cancer Beside organization.Organize clinical data.The corrected gene transcription profile data of 182 samples were substituted into the CIBERSORT algorithm,and the normalized gene expression data was used to infer the relative proportions of the 22 types of infiltrating immune cells in the sample.Because CIBERSORT uses Monte Carlo sampling,a P value can be derived for the deconvolution of each sample,so that the P value can be used to judge the accuracy of the prediction result.After deleting the data of P>0.05,138 samples were obtained,of which 134 were tumor tissues and 4 were paracancerous tissues.First,the Wilcoxon rank sum test or Kruskal-Wallis rank sum test was used to compare unpaired samples,and the differences in expression of 22 immune cells in 134 tumor tissues and 4adjacent tissues were analyzed,as well as the expression between tumor infiltrating immune cells Relevance.The data of tumor infiltrating immune cells of 134 tumor tissues were combined with clinical trait data of pancreatic cancer,and samples with incomplete clinical data were deleted.Correlation.Finally,the data of gene transcription profiles of134 samples were combined with the survival time and outcome in clinical traits.The samples were divided into high expression groups and low expression groups according to the median expression of immune cells,and each was analyzed by kaplan-Meier method.The relationship between immune cell expression and survival was tested by long-rank.Results:The expression of monocytes(P=0.025)and non-activated CD4~+memory T cells(P=0.040)in 22 immune cells was different in tumor tissues and adjacent tissues.Among them,monocytes were more expressed in adjacent tissues.High,unactivated CD4~+memory T cells were more highly expressed in tumor tissues.There were varying degrees of correlation between the 22 infiltrating immune cells in pancreatic cancer,and CD8~+T cells and M0 macrophages have a moderate intensity negative correlation(Pearson correlation coefficient-0.55).The correlation analysis between the expression of immune cells and clinical characteristics in pancreatic cancer showed that there was no correlation between the expression of immune cells in age and gender and the clinical characteristics of pancreatic cancer(P>0.05).The expression of follicular helper T cells was correlated with tumor grade(P=0.010),and the expression in G3 phase was higher than that in G2phase.Regarding the clinical stage of TNM,the expression of primitive B cells was correlated with T stage(P=0.044),and the expression in T1+T2 stage was higher than that in T3+T4 stage.No correlation was found between N stage and the expression of 22immune cells.The expression of activated mast cells,activated dendritic cells,activated CD4~+memory T cells was related to M stage,among which activated mast cells(P=0.003)and activated dendritic cells(P=0.046)The expression was higher in M1 than in M0,while activated CD4~+memory T cells(P=0.023)had higher expression in M0 than in M1.The expression of primitive B cells in 22 immune cells was related to clinical stage(P=0.045).Kaplan-Meier survival curve showed that high expression of CD8~+T cells(long-rank P=0.034)was associated with longer survival.Conclusion:The expression of monocytes and inactivated CD4~+memory T cells was different in pancreatic cancer tissues and adjacent tissues.There was a varying degree of correlation between the 22 immune cells in pancreatic cancer.immune cell expression was not correlated with age and gender,but correlated with tumor grade and TNM clinical stage.Immune cells associated with clinical traits of pancreatic cancer may have guiding significance for early diagnosis of pancreatic cancer patients.The expression of CD8~+T cells was correlated with the survival of pancreatic cancer.Detecting the expression of CD8~+T cells in patients with pancreatic cancer may be of guiding significance in judging the prognosis of patients with pancreatic cancer,and by increasing the infiltration of CD8~+T cells in pancreatic cancer tissues may help to increase the survival of patients with pancreatic cancer.
