Objective:This paper is designed to discusses the infulence of sodium pump alpha 2 subunits on myocardial ischemia reperfusion(ischemia reperfusion,IR)injury and further explore whether sodium pump signal pathways participate in myocardial ischemia-reperfusion injury through endoplasmic reticulum stress way.Methods:Constructed hypoxia reoxygenation model after the original generation culture of the myocardial cell.Each group was given different intervention during the hypoxia reoxygenation process.Experiments were divided into four groups:(1)inhibiting alpha 2 subunits sodium pump + IR group,(2)control + IR group(3)blank group,(4)hypoxia reoxygenation group(I/R).After completion of hypoxia reoxygenation,observe the cell growth situation by fluorescence microscope;Quantitative PCR to detect the sodium pump the?2 subunits mRNA expression;detecte apoptosis index through flow cytometry;and the expression levels of GRP78,endoplasmic reticulum stress signaling molecules and Bax,apoptosis protein were aslo detected by western blot.Results: 1,fluorescent quantitative PCR detection Sodium pump?2 subunits mRNA:compared with control group,the I/R group and the control-I/R group cell?2 subunits mRNA were decreased(P < 0.05);Compared with I/R group,the sodium pump ? 2si RNA-I/R group cell mRNA levels significantly lower(P < 0.05)2,The apoptosis index: Compared with the control group,the I/R group and the control-I/R group cell apoptosis rate was significantly higher(P < 0.05);Compared with I/R group,the sodium pump ?2 si RNA-I/R group,cell apoptosis rate increased significantly(P < 0.05)3,Western blot: Compared with control group,the control-I/R group CHOP in cells and GRP78 the amount of protein expression significantly higher(P < 0.05);Compared with I/R group,the sodium pump alpha?2 si RNA-I/R group of cells to CHOP and GRP 78 the amount of protein expression is also significantly higher(P < 0.05).Conclusion:1,Inhibition of sodium pump ? 2 subunit activity may aggravates endoplasmic reticulum stress,and increases myocardial cell apoptosis.2,Sodium pump alpha 2 may participate in myocardial ischemia-reperfusion injury through endoplasmic reticulum stress way.
