The Effects Of Xin-Yi Capsule On Endoplasmic Reticulum Stress Caused Cell Apoptosis In Myocardial Ischemia Reperfusion Injury Rats

Objective To observe the effects of Xin-Yi capsule on endoplasmic reticulum stress caused cell apoptosis in myocardial ischemia reperfusion injury rats, discussing its protective effect on myocardial ischemia-reperfusion injury and its mechanism of action.Methods150 healthy male SD rats were randomly divided into the sham group, the model group, compound Dan-Shen dropping pills(30 mg/kg), Xin-Yi capsule high, medium and low dose group(8.4 g/kg, 4.2 g/kg, 2.1 g/kg). Using ligation the left anterior descending coronary artery of rat heart, the method of myocardial ischemia-reperfusion model was set up. S-T segment change was recorded, Separation of serum, heart taken. the measurement of CK-MB?c Tn T?SOD?GSH-Px by ELISA,The histopathology of heart tissue was observed by HE staining, The expression of CHOP?Capase-12 m RNA was detected by RT-PCR, The expression of GRP78 protein was detected by WB, The apoptosis of cells in heart tissue was measured by TUNEL staining.Results1.Compared with model group, the experiment revealed that Xin-Yi Capsule attenuated S-T segment elevation(P <0.05?0.01).2. Compared with model group,the levels of CK-MB ? c Tn T decreased significantly in drug groups(P <0.01).3.Compared with model group,the levels of SOD ? GSH-PX was increased significantly in drug groups(P<0.01).4.Compared with sham group,myocardial fiber order, dense, did not see fracture,the structure is clear, the phenomenon of myocyte hypertrophy and atrophy, oval nucleus, are uniformly distributed in Model group; Compared with model group,visible muscle fiber fracture, disorder, myocardial cell gap widened, shrink phenomenon, which has a bleeding, nucleus pycnosis, burst; Xin-Yi capsule high dose group, middle dose group, low dose group and compound danshen dropping pill,visible muscle fiber arrangement is neat, dense, not seen obvious fracture, myocardial cells arranged relative rules, no obvious hypertrophy, edema in drug groups.5. Compared with model group, The expression of CHOP?Capase-12 m RNA was decreased in drug groups(P<0.01).6. Compared with model group, The expression of GRP78 protein was decreased in drug groups(P<0.01).7. Compared with model group, the apoptosis cells were decreased in drug groups by TUNEL(P<0.01).Conclusion Xinyi capsule may decrease the myocardial apoptosis cells,which be related with endoplasmic reticulum stress mediated apoptosis in myocardial ischemia-reperfusion injury in rats.
