Objective To investigate the effect of Chaihuang Weikuining Decoction on hypoxic microenvironment of gastric mucosa in gastric ulcer rats with liver constraint and spleen deficiency syndrome,by observing its effect on SOD,HIF-1a and LDH,and to explore its possible mechanism,clear the essence of gastric ulcer with liver constraint and spleen deficiency syndrome,to further promote the development of the clinical application of Chaihuang Weikuining Decoction.Methods 60 male SD rats were randomly divided into two groups:normal group(n = 20)and gastric ulcer rats with liver constraint and spleen deficiency syndrome group(n = 40).On the basis of acetic acid burning method,Construct the model of gastric ulcer rats with liver constraint and spleen deficiency syndrome by combination of various methods to simulation of TCM etiology.At the end of the model,10 rats were randomly selected from the normal group and the model group,which is used to verify the model.Then the remaining 30 model rats were randomly divided into gastric ulcer rats with liver constraint and spleen deficiency syndrome group,HQJZT group and CHWKN group,10 rats in each group.The remaining 10 rats in the normal group continued to be the control group.From the second day,HQJZT group and CHWKN group in the decoction by gavage for 14 days continuously.Each group was sacrificed at 24h after the last administration,and collect abdominal aorta blood as a sample.Use ELISA to detected the levels of serum superoxide dismutase(SOD)and hypoxia inducible factor-la(HIF-la)and lactate dehydrogenaseA(LDH-A).Results ? Compared with the normal group,the weight of the model group had significant difference(P<0.01);GAS,AMY and 5-HT were different degrees of lower than those in the normal group(P<0.05 or P<0.01);NE level was significantly higher than that in normal group(P<0.01);The color of the surface of the gastric mucosa was pale,with edema,exudate covering,endoscopic gastric mucosal layer defect,glands arranged in disorder,the main form of cells,parietal cells and mucous cells were irregular,visible inflammatory cell infiltration,thickening of the muscle.The model group met the requirements of gastric ulcer with liver constraint and spleen deficiency syndrome,indicating that the model was successfully constructed.? The HIF-1 a and LDH contents in the model group were higher,while the SOD content was low.After 14 days of treatment with Chaihuang Weikuining Decoction,the levels of HIF-la and LDH in the experimental group were lower than those before treatment,while SOD increased.Compared with the model group,the difference of SOD,LDH and HIF-la was statistically significant(P<0.05).Conclusion ? The expression of HIF-1a and LDH increased in the model group,while SOD showed low expression,which further confirmed the change of hypoxic microenvironment in gastric ulcer rats with liver constraint and spleen deficiency syndrome.? Through the change of HIF-la and LDH expression after drug intervention,it is clear that Chaihuang Weikuining Decoction can regulate the energy metabolism of three carboxylic acid cycle and improve the hypoxic microenvironment of gastric ulcer with liver constraint and spleen deficiency syndrome;Increase the expression of SOD,enhance the ability to scavenge oxygen free radicals and hypoxia tolerance,reduce the damage of lipid peroxidation to tissue cells.? Chaihuang Weikuining Decoction has the effect of improving the hypoxia of gastric ulcer rats with liver constraint and spleen deficiency syndrome and repairing gastric mucosa,and its mechanism may be related to the regulation of mitochondrial energy metabolism and the reduction of oxygen free radicals. |
