Objective To explore Chinese medicine Caesalpinia Sapan L and its effective components Brazilin on the protective effect and mechanism of vascular endothelial cell injury induced by high glucose,and provide experimental basis for the treatment of diabetic macrovascular disease.Methods Animal experiment:30 healthy male SD rats were randomly divided into normal group(n=10)and model group(n=20),normal group give normal feeding,model group give high fat and high glucose diet for 4 weeks,then inject of streptozotocin(STZ)solution one time,caused by diabetes model rats were randomly divided into control group and drug group.According to the human equivalent dose of Caesalpinia Sapan L group every day 810mg/kg content,that dissolved in 2ml distilled water to drug group,The normal group and the control group were given the same dose of normal saline every day.Also measure their blood glucose and body weight,take out aorta after 4weeks,using HE staining,transmission electron microscopy methods to discuss the effects of vascular endothelial cells autophagy by diabetes.Cell experiment:Cultivate human umbilical vein endothelial cells(HUVEC),determine the Brazilin concentration by MTT method,Adding glucose concent was 33.3 mmol/ l,the establishment of high glucose induced HUVEC damage in vitro model,given the concent of 15 ug/ml Brazilin.protein immunoblot assay autophagy related genes Beclin1,LC3-?,p62 m RNA and protein expression.Results Animal experiment:Diabetes building successful 18,just randomly divided into control group(n = 9)and drug group(n = 9),all live rats,normal color,more drinks,more food,polyuria and loss of weight.blood glucose as the experiment building front increased significantly(P < 0.05),the control group rats weight lower than the normal group,drug group was lower than the control group.Observation on the morphology of rat aortic HE staining,Caesalpinia Sapan L group and control group,compared with the normal group of vascular smooth muscle cells were damaged,but Caesalpinia Sapan L better than in the control group,although the was damaged,but has improved.To observe the autophagy structure of vascular endothelial cells in diabetic rats by transmission electron microscope,Caesalpinia Sapan L group compared with normal group,autophagosomes in number and volume increased;Mitochondrial cristae were disordered or vacuolated.Compared with the control group,the number of autophagy in diabetic rats was significantly lower than that in the control group.Conclusions Traditional Chinese medicine Caesalpinia Sapan L and Brazilin play a protective role on high glucose induced vascular endothelial cells by regulating autophagy,provide experimental basis for the treatment of diabetic macrovascular disease.
