Objective: To study the expression of SSB1 and the apoptosis in parotid gland tissue of rat with different doses of radiation and a radiation dose at different time points,which provid a new theoretical basis to further study about the radioactive damage mechanism in salivary gland.Methods: The rats were randomly divided into control group,6 groups with different doses of radiation,8 groups with a radiation dose at different time points,established a rat model of radiation and collected specimen;Tunel method is applied to detect Cell apoptosis after radiation;Immunohistochemical was used to detect the expression of SSB1 in rats parotid gland tissue in control and experimental group;RT-qPCR was applicated to detect the expression of SSB1 mRNA in rats parotid gland tissue in control and experimental group;WB method is applied to detect the expression of SSB1 in parotid gland tissue in control and experimental group.Results:The results of TUNEL show that the apoptosis rate increases with the radiation doses and it reached to the highest in group 12 Gy and then decreased(P<0.05).After the same 12 Gy radiation,the rate of apoptosis in the 1hour group was highest(P<0.05),and there was no obvious change at the later stage;The results of IHC,RT-qPCR,WB showed that SSB1 increases with the radiation doses,to a certain degree,its expression decreased after different doses of radiation(P<0.05).the expression of SSB1 were displayed a increasing first and then decreased after irradiation in a dose of 12Gy(P<0.05).Conclusions: Normal parotid gland tissues also can express a certain amount of SSB1;Radiation induced the expression of SSB1 in the parotid gland of rats;the expression of SSB1 was associated with the doses and times after radiation;Apoptosis is an early form of radiation damage in rat parotid gland,it is parallel with the expression rule of SSB1 in general,which showed the trend of increasing first and then decreasing;all of these provid a new theoretical basis to further study about the radioactive damage mechanism in salivary gland.
