Objective 1.To study the expression and distribution of SSB1 in normal parotid gland tissue in rat.2.To investigate the rule of SSB1 in rat' parotid gland tissue at different time after radiation,as well as the repair of DSB damage,and explore the relationship between SSB1 and radioactive damage repair.Methods 1.Take the normal rats(n=5)as control,the experimental rats(n=40)were radiated with60 Co gamma rays on head and neck region in a dose of 6 Gy.2.Immunohistochemical was used to detect the expression of SSB1 and?-H2AX in rats parotid gland tissue in control and experimental group at different time points after radiation.3.RT-qPCR was applicated to detect the expression of SSB1 mRNA in rats parotid gland tissue in control and experimental group at different time points after radiation.Results 1.A certain amount of SSB1 was detected in part of acinar cells and all duct cells in parotid gland tissue in control group,located in the nucleus,and was not found in connective tissue.2.The results of immunohistochemical and RT-qPCR showed that SSB1 were displayed a increasing first(P<0.05)and after weakening trend over time after irradiation in a dose of 6 Gy,and the unper expression was close or lower to nomal.3.The result of immunohistochemical showed that a small amount of?-H2AX was also detected in normal parotid gland cells,located in the nucleus.Its expression increased significantly(P<0.01),reaching peak at 1 hour,and dropped close to normal levels in the later after radiation in a dose of 6 Gy.Conclusions SSB1 is a radiation-induced protein and may be involved in radioactive damage repair process in the rats' parotid gland,providing a new theoretical basis to further study the salivary gland radioactive damage mechanism.
