Objective: To study the expression level of DNA methyltransferase 1(DNMT1)and DNA methyltransferase 3A(DNMT3A)and DNA methyltransferase 3B(DNMT3B)mRNA and protein in patients with primary gout.Methods: Real-time quantitative polymerase chain reaction(RT-qPCR)was employed to measure the expression of DNMT1,DNMT3 A and DNMT3 B m RNA in peripheral blood mononuclear cells(PBMCs).The expression of DNMT1,DNMT3 A and DNMT3 B gene mRNA in PBMCs were compared among patients with acute gout patients(AG,n=50),intermediate gout patients(IG,n=47)and healthy controls(HC,n=44),and the otherness of gene expression in three groups were compared.Western blot(WB)was used to study the protein expression of DNMT1,DNMT3 A and DNMT3 B in PBMCs.All features were analyzed by SPSS 19.0 statistical software,Wilcoxon test,One-Way analysis of variance was used for statistical analysis.Results: The expression of DNMT1 mRNA in the group of AG and IG[0.0008(0.0004,0.0051),0.0011(0.0004,0.0084)]were significantly lower than that in the HC group [0.0017(0.0010,0.0083),F=3.696,P<0.05],The expression difference of DNMT1 mRNA between the group of AG and IG was not statistically significant(P > 0.05),The expression of DNMT1 protein in the group of AG and IG[0.7101(1.1141),0.7489(1.7913)]were significantly lower than that in the HC group [1.7009(5.8011),F=4.41,P<0.05)],The expression difference of DNMT1 mRNA and protein between the group of AG and IG was not statistically significant(P > 0.05).The Expression of DNMT3 A mRNA in the group of AG and IG[0.0058(0.0015,0.0179),0.0128(0.0032,0.0435)] were significantly lower than that in the HC group [0.0151(0.0076,0.0727)],F=10.145,P<0.05],The expression of DNMT3 A protein in the group of AG and IG[0.3301(0.1802),0.5611(0.2597)] was significantly lower than that in the HC group [0.6719(0.1599),F=18.68,P<0.05],The expression difference of DNMT3 A mRNA and protein in the group of AG was significantly lower than that in the IG group,It was statistically significant(P <0.05).The expression of DNMT3 B mRNA in the group of AG ? IG and HC were no statistically significant [F=1.86,P>0.05].The expression level of DNMT1 was correlated with Crea(r=0.394,P<0.05),The expression level of DNMT3 A was correlated with MCV(r=0.588,P<0.05).Conclusions: The abnormal expression of the DNMT1,DNMT3 A and DNMT3 B mRNA and protein in primary gout patients,suggest that they were involved in the pathogenesis of gout.
