| Objectives: To explore the role and the clinical significance of the tumor suppressor gene INPP4 B and PTEN in nodular thyroid disease organizations by detecting the expression of both of them in normal thyroid tissue and nodular thyroid disease organizations,combined with the clinical pathological features.To look for new tumor markers for thyroid cancer and provide a theoretical basis for developing the corresponding molecular targeted drugs.Method: Collecting 134 effective samples of nodular thyroid disease from July 1,2014 to June 1,2016 in north Sichuan medical college affiliated hospital,which were diagnosed in Endocrinology department,including 41 cases of nodular goiter,45 cases of thyroid adenoma,48 cases of thyroid carcinoma(43 cases of papillary carcinoma,3 cases of follicular carcinoma,1case of undifferentiated carcinoma,1 case of medullary carcinoma),20 cases of normal thyroid tissue.Using immune histochemical method to detect the expression of tumor suppressor gene INPP4 B and PTEN.Using SPSS13.0statistical software for data analysis,inspection on both sides,α=0.05 as the inspection level,if p<0.05 the difference was statistically significant.The relation of the expression of two genes in different pathological types of thyroid nodule was analyzed by the spearman rank correlation analysis.The chi-square test was used in the comparison of genes expression in normal tissues,nodular goiter,thyroid adenoma,thyroid carcinoma,as well as the different clinical pathological characteristics.Results:(1)In thyroid carcinoma tissue,thyroid adenoma tissue,nodular goiter,normal tissue,INPP4 B gene protein expression had increasing trend,but INPP4 B gene protein expression in normal tissue,nodular goiter tissue,thyroid adenoma tissue did not have significant difference(p>0.05).The protein expression level of INPP4 B gene in thyroid carcinoma tissue was significantly lower than those in benign nodular thyroid disease tissues,the difference was statistically significant(p<0.05).(2)In thyroid carcinoma tissue,thyroid adenoma tissue,nodular goiter,normal tissue,PTEN gene protein expression had increasing trend,but PTEN gene protein expression in normal tissue,nodular goiter tissue,thyroid adenoma tissue did not have significant difference(p>0.05).The protein expression level of PTEN gene in thyroid carcinoma tissue was significantly lower than those in benign nodular thyroid disease tissues,the difference was statistically significant(p<0.05).(3)In nodular goiter tissue and thyroid adenoma tissue,the INPP4 B and PTEN gene protein expression was not statistically correlated with age,gender,the size of nodule and calcifications(p>0.05).(4)In thyroid carcinoma tissue,the INPP4 B gene protein expression was statistically correlated with gender,the size of nodule,calcifications,neck lymph node metastasis.The protein expression level of INPP4 B gene in female patients,>2cm tubers,calcifications,neck lymph node metastasis was significantly lower than those in male patients,≤2cm tubers,no calcification,no neck lymph node metastasis,the difference was statistically significant(p<0.05),no correlation with age(p>0.05).(5)In thyroid carcinoma tissue,the PTEN gene protein expression was statistically correlated with calcification and neck lymph node metastasis.Theprotein expression level of PTEN gene in calcification and neck lymph node metastasis was significantly lower than those in no calcification and no neck lymph node metastasis,the difference was statistically significant(p<0.05).There was no correlation with age,gender,the size of nodule.(p>0.05).(6)In nodular goiter tissue,there was positive correlation between INPP4 B gene protein expression and PTEN protein expression(rs=0.396,p<0.05).In thyroid adenoma tissue,there was positive correlation between INPP4 B gene protein expression and PTEN protein expression(rs=0.448,p<0.05).In thyroid carcinoma tissue,there was positive correlation between INPP4 B gene protein expression and PTEN protein expression(rs=0.904,p<0.05).Conclusion:(1)The protein expression level of INPP4 B gene and PTEN gene in thyroid carcinoma tissue,thyroid adenoma tissue,nodular goiter tissue and normal thyroid tissue increased in turn.(2)The protein expression level of INPP4 B gene and PTEN gene significantly decreased in thyroid carcinoma tissue,which could be as a valuable reference in differential diagnosis of benign and malignant thyroid nodule.(3)The down of gene protein expression of INPP4 B and PTEN was closely related to the occurrence and development of thyroid carcinoma,as well as neck lymph node metastasis.(4)There were correlation and synergy between INPP4 B gene and PTEN gene in the occurrence and development of thyroid carcinoma.(5)The protein expression of INPP4 B gene was related to gender,which could be one of the reasons for difference of thyroid cancer incidence between men and women. |
