Objective To explore the effect of let-7c-1 influenced the learning and memory of PTZ-induced epileptic rats.Investigated the role of let-7c-1 in the cognitive function of PTZ-induced epileptic rats.Methods A model of temporal lobe epilepsy(TLE)was induced via PTZ kindling in SD male rats.The epileptic rats were divided into epilepsy group,agomir-control group,let-7c-1 agomir group,12 rats for each.12 rats were used as negative control group.After 28 days.Observe the behavior of the rats,the expression of let-7c-1,the expression of Bcl-2 protein and Caspase3 protein.Results 1.We found that compared to the negative group,the escape latency of epilepsy group was prolonged,the crossing times and the quadrant total distance in the target was reduced(P<0.05).compared to the epilepsy group and the agmoir-control group,they didn't have obviously differences(P>0.05).compared to agmoir-control group,the escape latency let-7c-1 agomir group was prolonged,the crossing times and the quadrant total distance in the target was reduced(P<0.05).2.Compared to the negative group,the expression of let-7c-1 in epilepsy group was higher(P<0.05).the expression of let-7c-1 in let-7c-1agomir group were higher than other group(P<0.05).3.Compared to the negative group,the expressions of Bcl-2 protein in other groups were decreased(P<0.05),the Caspase3 protein were increased(P<0.05).Compared to the Agomir-control group,the expression of Bcl-2protein in let-7c-1 agomir group was decreased obviously(P<0.05),and the expression of Caspase3 protein was increased(P<0.05).Conclusions1.It revealed that let-7c-1 may make worse in impairing the learning and memory of PTZ-induced epileptic rats.2.let-7c-1 could decrease the Bcl-2 protein and increased Caspase3 protein in the hippocampus that might be the way to impair cognitive function.
