Objective:In this dissertation,the studies were included the isolation and structure identification system on Aralia chinensis of triterpenoid saponins,as well as the quality standard of Anhui Aralia medicinall materials.Method for the determination ofharmful heavy metals content establishment of Aralia medicinal herbs,to evaluate the safety of Aralia medicinal naterials.Established the UPLC-ELSD method to determine the main active ingredients like congmujingnosideA,congmujingnosideB and congmujingnosideC which were isolated from the stem of Aralia chinensis and Aralia echinocaulis.Methods:1.The compounds were isolated and purified from the stems of Aralia chinensis by chromatography packed with ODS,HPLC and their structures were identified by means of spectral data analysis.2.Pharmacognosy identification of the sample include the plant morphology,medicinal herbs traits,microscopic characteristics and TLC identification.3.The sample pretreatment method was microwave digestion.Then heavy metal contents were determined by ICP-MS.4.To determine the contents of the stem of Aralia chinensis and Aralia echnocaulis.Use the UPLC-ELSD with the ACQUITY UPLC(?)Phenyl(2.1 × 1 00 mm,1.7 ?m)column.And the mobile phase composed of acetonitrile-water(26:74),the flow rate was 0.2 ml/min,and the temperature of column was 35 ?,atomization temperature was 45?,the drift tube temperature was 80?,the carrier gas pressure of 20 psi and the gain value is set to 20.Results:1.Nine compounds were isolated from the butyl alcohol extraction part of stem of Aralia chinensis and Aralia echinocaulis.On the basis of chemical and spectral evidence,there were seven new compounds,their structures were determined as follows:congmujingnoside A(3-0-{[?-D-glucopyranosyl-(1?2)]-[?-D-glucopyrano-syl-(1?6)-?-D-glucopyranosyl-(1?3)]-?-D-glucopyranosyl}oleanolic acid 28-O-?-D-glucopyranosyl ester)(1),congmujingnoside B(3-0-{[?-D-xylopyranosyl-(1?2)]-[?-D-glucopyranosyl-(1?6),?-D-glucopyranosyl-(1?3)]-)?-D-glucopyranosy l}oleanolic acid 28-O-?-D-glucopy ranosyl ester)(2),congmujingnoside C([?-D-xylopyranosyl-(1?>2)]-{[?-D-glucopyranosyl-(1?3)]-[?-D-glucopyranosyl-(1?6)]-?-D-glucopyranosyl-(1?3)}28-0-?-D-glu copyranosyl oleanolic acid ester)(3),congmujingnoside D([?-D-glucopyranosyl-(1?2)]-{[?-D-glucopyranosyl-(1 ?3)]-?-D-glucopyranosyl-(1?3)}28-O-?-D-glucopyranosyl oleanolic acid ester)(6),congmujingnoside G([?-D-xylopyranosyl-(1?2)]-{[?-D-quinovopyranosyl-(1?6)]-?-D-glucopyranosyl-(1?3)}28-O-?-D-glucopyranosyl oleanolic acid ester)(9).And two compounds were first isolated from this plant,their structures were determined as follows:Congmunoside V(4)and Araliasaponin IV(5).2.The systematic study were quality standard of Aralia chinensis and Aralia echinocaulis were studied,developed a method to evaluate the quality of Aralia medicinal materials.The source of two kinds of medicinal properties,microscopic characteristics,TLC identification and other aspects of the detailed pharmacognostic identification,which determined the identification basis of Aralia taibaiensis.It can provide a scientific basis for the identification and utilization of the medicinal materials.3.A method that is ICP with microwave digestion was established for determination of the contents of five kinds of heavy metals.The linear relationship of each element was good(r=0.991-0.999),the average recovery ranged from 88.7%-101.2%.According to the Green standards of medicinal plants and preparations for foreign trade and economy,5 kinds of heavy metal content in the 10 batches of Aralia chinensis did not exceed the standard.Only 6 batches of Aralia echinocaulis chromium content exceed the standard.4.The linear relationship of congmujingnosideA which ranged from 0.04?1.20 ?g was good.The average recovery was 101.0%,RSD was 1.89%.The linear relationship of congmujingnosideB which ranged from 0.25-1.00 ?g was good.The average recovery was 98.1%,RSD was 1.63%.The linear relationship of congmujingnosideC which ranged from 0.23?1.00 ?g was good.The average recovery was 97.8%,RSD was 2.49%.The average content of 10 batches of congmujingnosideA was 0.190%,congmujingnosideB was 1.356%and congmujingnosideC was 0.417%.Consults:9 compounds were obtained from the butyl alcohol extraction part of stem of Aralia chinensis.Among them,compounds1?2?3?6?7?8?9 were new compounds.For the first time,Compounds 4,5 were isolated from this plant.Origin identification,macroscopic identification,physicochemical identification,microscopic identification can be used for systematic identification of Aralia plants in Anhui province as well as providing a new method for accurate,rapid identification of Aralia.A method that is ICP with microwave digestion was established for determination of the contents of five kinds of heavy metals.The linear relationship of each element was good(r=0.991-0.999),and the average recovery ranged from 88.7%-101.2%.According to the Green standards of medicinal plants and preparations for foreign trade and economy,only the Aralia echinocaulis chromium content exceed the standard.To develop the analytical method to determine the content of congmujingnosideA?B?C in the stems of Aralia chinensis and Aralia echinocaulis by UPLC-ELSD.The method was convenient,reliable and with good reappearance.
