Objective Atopic dermatitis(AD)is a chronic inflammatory skin disease associated with atopy,It's manifestations is itching,multiple lesions and a tendency to exudation,often associated with asthma,allergic dermatitis.The main methods of treating AD is external use glucocorticoid and calcineurin inhibitor.But its long-term use will produce many adverse reactions.And the latter is expensive,aggravate the economic burden of patients.More importantly,we found in clinical observation: both of them have good effect on AD acute lesions,and they have Poor effect on Subacute and chronic lesions.We are in urgent need of an effective,economical and safe drug for the treatment of subacute and chronic skin lesions.In this study,AD mice model was established by DNCB.After the modeling success,Compound cream of Baiji group,Cream group,Cetaphil group were respectively using Compound cream of Baiji,Cream,Cetaphil moisturizing lotion.Treatment lasted twoweeks.After treatment,detect each group's IgE?IL-4?IFN-? level,and Observe each group's skin pathological condition.Whether Compound cream of Baiji effective confirmed by this experiment.And explain how it works.Method1)Animal grouping method:50 mice were divided into 5 groups:Blank group,Model group,Compound cream of Baiji group,Cream group,Cetaphil group.2)Method for making model : Shave off the back hair of mice,Area is about2cm×2cm.Acetone and olive oil mixed by 3: 1,Configure DNFB to 0.5%concentration.The first and second day use 0.5%DNFB inunction mice back skin.3-6days without inunction.Starting from the seventh day inunction interval once a day.28 th days to observe the skin of mice,If appear erythema and desquamationi,this indicate success.3)Intervention method:Compound cream of Baiji group,Cream group,Cetaphilgroup were respectively using Compound cream of Baiji,Cream,Cetaphil moisturizing lotion.Once a day,One time 100?l.Blank group not treated,Model group:Inunction mixed solution,One time 100?l,Once a day.Treatment lasted two weeks.4)Index determination and method:(1)Histopathological examination 14 th days kill mice,Get back hair removal skin,Fixed with 10% Formaldehyde Solution,paraffin embedding,section,after HE staining,observe pathological changes by microscope.(2)Testing levels of IL-4,IFN-?,and IgE:Using the ELISA method?5)Statistical method Data representation method:Mean±standard deviation(sx ?),Statistical analysis using SPSS17.0 software.Single factor analysis of variance was used for multiple samples.Homogenety of test was used to test the homogeneity of variance,If the variance is not uniform,dunnerte method was used for comparison.If the variance is uniform,Bonferroni method was used for comparison.Result1)Expression level of IgE in each group The level of IgE in the model group was significantly higher than that in the blank group,the difference was statistically significant,(P=0.000).The level of IgE in the Compound cream of Baiji group was lower than that in the model group.the difference was statistically significant,(P=0.000).The level of IgE in the Cream group was lower than that in the model group.the difference was statistically significant,(P=0.003).The level of IgE in the Cetaphil group was lower than that in the model group.the difference was statistically significant,(P=0.001).2)Expression level of IL-4 in each group The level of IL-4 in the model group was significantly higher than that in the blank group,the difference was statistically significant,(P=0.014).The level of IL-4 in the Compound cream of Baiji group was lower than that in the model group.the difference was statistically significant,(P=0.000).The level of IL-4 in the Cream group was lowerthan that in the model group.the difference was not statistically significant,(P=0.899).The level of IL-4 in the Cetaphil group was lower than that in the model group.the difference was statistically significant,(P=0.000).3)Expression level of IFN-? in each group The level of IFN-? in the model group was significantly lower than that in the blank group,the difference was statistically significant,(P=0.000).The level of IFN-? in the Compound cream of Baiji group was higher than that in the model group.the difference was statistically significant,(P=0.020).The level of IFN-? in the Cream group was higher than that in the model group.the difference was not statistically significant,(P=0.595).The level of IFN-? in the Cetaphil group was higher than that in the model group.the difference was not statistically significant,(P=0.639).4)Severity comparison of each group mice:Blank group :No obvious change Model group:Skin back have erythema infiltration,covering scab.Same as eczema skin.Cream group:Skin like model group and have a little better.Compound cream of Baiji group:Skin lesions relieved significantly.Leave a little red spot.Cetaphil group:Skin lesions relieved significantly.More than the Compound cream of Baiji group of erythema.5)Comparison of histopathology of skin1.Blank group :No obvious change.2.Model group:Hyperkeratosis,spinous layer hypertrophy,sponge edema,dermal vascular expand and congestion,a large number of lymphocyte eosinophilic granulocyte in dermis.3.Compound cream of Baiji group:Slight hyperkeratosis,prickie cell thinner than model group,mild inflammatory cell infiltration4.Cream group:Pathological performance was slightly reduced compared with model group.5.Cetaphil group:Compared with compound cream of Baij group,inflammatory cell infiltration is more serious,prickie cell hypertrophy than compound cream of Baij group.Conclusion1.Compound cream of Baiji can reduce AD mouse model IgE and IL-4 level.Rise AD mouse model IFN-? level.2.Compound cream of Baij can cure AD mouse mode skin lesions.
