Objective:To detect the plasma levels of immunoglobulin E (IgE), interlukin17(IL-17) and interlukin23(IL-23) in the Nc/Nga mouse atopic dermatitis(AD) model, and to explore the treatment effect of Rutaecarpin cream in Nc/Nga mouse atopic dermatitis model and its possible mechanism.Method70Nc/Nga mice were randomly divided into atopic dermatitis model group and acetone normal control group. In the model group, we used5%2,4-nitro chlorobenzene (DNCB) to repeatedly stimulate the skin surface of mice back to build___atopic dermatitis model,acetone for control group. Then the50mice in model group were divided into five different sub-groups (10mice per group):model natural recovery group, rutaecarpin-treated groups of high dose, middle dose and low dose,0.075%dexamethasone acetate cream-treated positive control group. We observed the changes of skin lesion and its lesion histopathology and detected the serum levels of IgE, IL-17and IL-23in mice of different groups.Result:Compared to acetone-treated normal control group, AD model mice control group presented with typical eczema dermatitis, it’s histopathogical results showed erosion and crust of epidermis, spongiosis in stratum spinosum layer, hemangiectasis and lymphocytes infiltration in dermis. There was a statistically significant increase of serum levels of IgE, IL-17, IL-23in mice of model groups compared to acetone-treated control group.Compared to natural recovery group of AD model,the dermatitis degree and histopathological changes were significantly improved in rutaecarpin-treated groups of different doses, especially in high dose and middle dose groups, which almost had the same effect as that being seen in0.075%dexamethasone-treated group. The plasma levels of IgE, IL-17, IL-23had a statistically significant decrease in mice of rutaecarpin-treated groups, especially in high dose and middle dose groups, which was similar to that of dexamethasone cream-treated group. Compared with acetone treated normal control group, the dermatitis degree and histopathological results were relatively improved in model natural recovery group, and the serum levels of IgE, IL-17, IL-23decreased.Conclusion:The DNCB induced Nc/Nga mice AD model showed the same changes in Thl7lineage IL-23/IL-17axis as that in human AD. Rutaecarpin improved the recovery of AD-like Nc/Nga model probably by inhibiting the expression of IL-17and IL-23anddecresing the IgE level.
