Objective: 1 To obtain the best polar parts of Antioxidant Activity of Xinfeng Capsule,and conduct qualitative and quantitative research on its Antioxidant components.2 To establish the TLC spectrum of two antioxidant components of Xinfeng Capsule in order to improve the quality control standard.3 Make up the gap of both clinical research and pharmaceutical research to provide more basis for clinical application,for the well-being of patients.Methods:1 Antioxidant activity evaluation system is established to compare antioxidant activity of different polar parts to determine the composition of the active parts through a variety of antioxidant capacity in vitro detection methods.Evaluating the free radical scavenging capacity and total reducing capacity of two parts of Xinfeng Capsule to parallel comparison of their antioxidant activity.Using DPPH,ABTS,Tris–HCl,Fenton and total reduction capacity.And extend to make determination of total flavonoids and total phenols in different polar parts to explore the antioxidant composition of them.Thin Layer Chromatography was used to investigate the antioxidant components in the best polar part to track the effective substance basis of antioxidant activity.High Performance Liquid Chromatography(HPLC)was used to further identify the antioxidant components in the best polar part and determe the content.2 To take a TLC-DPPH radical scavenging bioautography technique based on two of them used to identify flavonoids antioxidant components.3 Combined with a variety of methods,including the determination of antioxidant capacity in vitro,the qualitative and quantitative analysis of antioxidant components,and DPPH-TLC mapping,the first step of the study on the antioxidant activity of Xinfeng capsule was made.Results:1 Ethyl acetate extract of Xinfeng capsule has good antioxidant activity,comparing with other polar parts of antioxidant components.In ethyl acetate polar part of Xinfeng Capsule,there are three antioxidant components including calycosin,calycosin-7-O-?-D-gly-coside and formononetin.Among them,each 1 g Xin Feng capsule contains 0.0222×10-4% calycosin-7-O-?-D-gly-coside,0.0266×10-5% calycosin,0.0149×10-5% formononetin.At the same time,Triptolide was not detected,consistented with the results of the TLC study.2 Establishment of TLC spectrum of two antioxidant components(calycosin and calycosin-7-O-?-D-gly-coside)of Xinfeng Capsule Successfully to improve the quality control standards.3 In this study,through the preliminary study on the antioxidant activity of Xinfeng capsule,the clinical research and pharmaceutical research of the antioxidant activity were connected,which provided more evidence for clinical application.Conclusion: Xinfeng Capsule has good antioxidant activity.In the polar parts of Xinfeng Capsule,the polar part of ethyl acetate is rich in many kinds of antioxidant components and shows the strongest antioxidant activity.It can be used as a new natural antioxidant.calycosin,calycosin-7-O-?-D-gly-coside and formononetin are antioxidant constituents in Xin Feng Capsule.Therefore,we speculate that flavonoids may be the main material basis of antioxidant effect of Xinfeng capsule.This study provides a relatively simple and systematic method and thinking for the research of antioxidant activity in the hospital.
