ObjectiveSupplementing qi and activating blood circulation method is one of the therapeutic principles in atherosclerosis(AS).In order to study the therapeutic effect and the mechanism in AS by using supplementing qi and activating blood circulation method,This research based on AS ApoE-/-mice model which established by feeding mice with high-fat diet,then Buyang Huanwu Added Decoction was chosen to intervene.At length,providing a scientific theoretical basis and experimental evidence for treating atherosclerosis by using Supplementing Qi and Activating Blood Circulation Method.on formation of atherosclerosis.Methods30 Male C57BL/6J mice and 30 male ApoE-/-mice were chose,both of them were 7-8 week-age.C57BL/6J mice was as a control group,and male ApoE-/-mice were randomly divided into four groups:ApoE-/-mice control group,model group,Buyang Huanwu Added Decoction group and simvastatin group.Model group,Buyang Huanwu Added Decoction group and simvastatin group were established by high-fat diet.4 weeks later,Buyang Huanwu Added Decoction group and simvastatin group were intragastrically administrated with Buyang Huanwu Decoction and leech powder and simvastatin.After 9 weeks of treatment,cholesterol(TC),triglyceride(TG),high density lipoprotein cholesterol(HDL-C)and low density lipoprotein cholesterol(LDL-C)were detected by automatic biochemical analyzer.In addition,aorta segment was taken from the experiment animals to observe morphological changes by Hematoxylin and eosin(HE),also measure fiber-cap thickness and vascular intima-media thickness.Westem-blot was used to detect the expression of p38 mitogen-activated protein kinase(p38MAPK)and extracellular-regulated kinase 5(ERK5)in mice aorta segment,it is explored that the therapeutic mechanism of Buyang Huanwu Added Decoction on formation of atherosclerosis in mitogen-activated protein kinase(MAPK)signal pathway.Results In blood lipid levels,compared with the normal group,TC,TG and LDL-C in the ApoE-/-mice control group and the model group were significantly higher,while HDL-C levels was significantly lower(P<0.05).Compared with the model group,TC,TG and LDL-C in the Buyang Huanwu Added Decoction group and simvastatin group were significantly lower,while HDL-C were significantly higher(P<0.05,P<0.05).Compared with the Buyang Huanwu Added Decoction group,TC and LDL-C in simvastatin group were significantlylower(P<0.05,P<0.05),while the changes in TG and HDL-C level were not significant(P>0.05).HE staining showed no aortic plaque formation in C57BL/6J mice control group,few aortic plaque formation and slightly increased vascular intima-media thickness in ApoE-/-ce control group,significantly aortic plaque formation and increased vascular intima-media thickness in model group.Compared with the model group,aortic plaque formation and vascular intima-media thickness could be reduced by both Buyang Huanwu Added Decoction group and simvastatin group.Compared with the Buyang Huanwu Added Decoction group,both aortic plaque formation and vascular intima-media thickness could be reduced in simvastatin group insignificantly(P>0.05),The expression of aortic p38 MAPK obviously increased while ERK5 decreased in model group(P<0.05,P<0.05),compared with C57BL/6J mice control group.And the expression of aortic p38 MAPK significantly decreased while ERK5 increased in both Buyang Huanwu Added Decoction group and simvastatin group,compared with model group(P<0.05,P<0.05).Compared with the Buyang Huanwu Added Decoction group,the expression of p38 MAPK significantly decreased(P<0.05),while ERK5 increased insignificantly(P>0.05)in simvastatin group.Conclusion1.Both Buyang Huanwu Added Decoction and simvastatin has a therapeutic effect in the development of atherosclerosis.2.The potential mechanism could be related to intervene of p38 MAPK and ERK5 in MAPK signal pathway.3.Providing an experimental evidence for treating atherosclerosis by using Supplementing Qi and Activating Blood Circulation Method.
