Objective: To observe the effect of ZhiGe Oral Liquid on the model rats with alcoholic liver disease(ALD)and its effect on related factors like lipid metabolism,immune mechanism,oxidative stress and ethanol metabolism,and to explore the preventive and therapeutic effect of this Oral Solution on ALD and its function mechanism.Methods: 1.Animal in groups: 85 SD male rats were randomly divided into 6 groups: normal group,model group of ALD animal(model group),high dose of ZhiGe Oral Solution group(high-dose group),middle dose of ZhiGe Oral Solution group(middle-dose group),low dose of ZhiGe Oral Solution group(low-dose group)and positive control group of Hangover Helper(control group).Normal group consists of 10 rats,while the remaining 15 per group.2.Molding and administration: Except for normal group,the other groups were given 52% Luzhou Laobaigan(produced by Luzhou Laojiao Co.,Ltd.)via gavage of 1.0ml · 100g-1 · d,and meanwhile,the high-dose group,the middle-dose group,and low-dose group were respectively fed with ZhiGe Oral Solution 1.0ml·100g-1·d,0.5ml·100g-1·d and 0.25ml·100g-1·d,and the control group was fed with Hangover Helper 0.5ml·100g-1·d,the normal group was fed with distilled water 1.0ml · 100g-1 · d.The above groups are free to water,and fed by normal diet.After 8 pm every day,the food was forbidden,but waterstill accessible.Diet was provided after gavage at 9:00 of the next day.Weight of the rats was recorded every Friday.All the rats were sacrificed at the end of the 12 th week while the serum and intact liver tissue were collected from each rat,and the wet weight of liver tissue was also measured.3.Target Parameters:HE staining was used to observe the structural changes of liver tissue.the Oil red O staining was used to explore the liver lipid condition.The serum levels of ALT,AST,TC and TG were measured by automatic biochemical analyzer.The contents of TC and TG in the liver were measured by colorimetric assay.The protein expression of the cytochrome of the liver was detected by Western Blot.TNF-?,IL-6,IL-10,ADH and ALDH were detected by enzyme-linked immunosorbent assay(ELISA).Results: 1.HE staining of liver tissue showed that the size of the hepatic lobules and the size of the cells in the normal group were uniform,the boundary was clear,which showed a neat and regular radial arrangement.The hepatic lobules of the rats in the model group had irregular structure,and the boundary was blur,and a vast area of fat vacuoles can be seen in cells,and part of nuclei deviated.The hepatocyte structure in the high-dose group was similar to that in the normal group.Compared with the model group,the middle-dose group and the control group were both improved.The model group and low-dose group were basically similar.2.the Oil red O staining showed that a very small amount of red fat particles can be seen in the part of the liver in the normal group.In the model group,a large area of liver cells can be seen,around which there are many red fat particles;the red lipid droplets of high-dose groupwere similar to that of the normal group;the middle-dose group and the control group were increased;while the low-dose group can see a large area of red fat particles gathered.3.Changes of liver index,serum IL-6,TNF-?,ALT,AST,TC,TG,ADH and Liver tissue of TC,TG and P4502E1 in each group: compared with the normal group,the model group were significantly higher(P<0.05).Compared with the model group,the high-dose group,the middle-dose group and the low-dose group presents an increasing trend,while the high-dose group was obviously lower than the model group(P<0.05),and the serum TG of the high-dose group was higher than that in the normal group(P<0.05).Other indexes of the high-dose group were similar to those of the normal group and there was no significant difference between the two groups(P>0.05).The control group was higher than that the middle-dose group,and there was no significant difference between these two groups(P> 0.05).4.Changes of Serum IL-10,ALDH: Compared with the normal group,the IL-10 and ALDH in the model group were lower than those in the normal group(P <0.05).The IL-10 in the high-dose group was lower than that in the normal group(P <0.05).The ALDH of the high-dose group was lower than that of the normal group,but there was no significant difference between the two groups(P> 0.05).Compared with the model group,the IL-10 and ALDH in the high-dose group,the middle-dose group and low-dose group were in an decreasing trend,and the high-dose group was significantly higher than model group(P <0.05),the IL-10 of control group was higher than that of the minddle-dose group(P> 0.05),but there was no significant difference between these two groups(P> 0.05).TheALDH of the Hangover Helper control group was lower than that of the middle-dose group,and there was no significant difference between these two groups(P> 0.05).Conclusion: 1.Normal diet and gradient alcohol gavage method can make the accumulation of fat in the liver of SD rats.Liver indexes of rats were increased,while the liver function damaged,blood lipids increased,and serum inflammatory factors increased,which contribute to the success of ALD modeling.2.ZhiGe Oral Solution can alleviate liver function injury,decrease liver index and blood lipids,reduce inflammatory cytokines in serum but increase anti-inflammatory cytokines,lower ADH content in serum while increase the ALDH content,and reduce the expression of hepatocyte P4502E1.thus its effect on alcoholic liver disease has a good prevention and cure.3.The function mechanism of ZhiGe Oral Solution's preventing and treating on alcoholic liver disease may be as follows:(1)it reduces lipid accumulation and lipid metabolism in liver tissues;(2)participates in immune regulation;(3)accelerates the metabolic rate of alcohol in the body,so as to delay the progress of ALD;(4)participates in the regulation of oxidative stress in the liver.
