This article was conducted to evaluate the effects of different oils on the antioxidation capability and blood lipid metabolism of mice by introgastricing(ig) different oils and feeding high-fat diet with different oils.Fifty-six male KM mice were divided into 7 groups: ig normal saline, coconut oil, lard oil, tea oil,oxidated tea oil,oxidated tea oil+Lipoic acid(LA)(0.05%),oxidated tea oil+LA(0.1%).The free radical(FR) of blood was detected by luminol Chemiluminescence method. Fifty-six male KM mice were divided into 7 groups fed with different diets:normal diet; coconut oil diet; lard oil diet; tea oil diet; oxidated tea oil diet; oxidated tea oil+LA(0.05%) diet; oxidated tea oil+LA(0.1%) diet. After 6 weeks, the level of FR, the content of malondialdehyde(MDA) and total antioxidative capacity(T-AOC) level, the activities of superoxide dismutase(SOD), glutathione peroxidase(GSH-Px) and catalase(CAT) in serum and tissues were measured respectively; the contents of triglyceride(TG), total cholesterol(TC), high density lipoprotein cholesterol(HDL-C) , low density lipoprotein cholesterol(LDL-C), the atherogenic index (AI), the activities of lipoprotein lipase(LPL)and hepatic lipase(HL) in serum and liver were also determined respectively. The expression of mRNA of somatostatin(SS), carnitine palmitoyl transferase-1(CPT-1), apolipoproteinE(ApoE) in liver or intestinal were detected by fluorescence quantify polymerase chain reaction(FQ-PCR) respectively. The results were shown as follows:1 After introgastricing lard oil, the level of FR was significantly higher than that of introgastricing tea oil and coconut oil(P<0.05).After introgastricing oxidated tea oil, the level of FR was significantly higher than that of normal(P<0.05); the level of FR can be decreased when LA is added into oxidated tea oil, and 0.1% is suitable.2. The high-fat diet increased the level of FR in blood ,decreased the T-AOC; decreased the activities of SOD, GSH-Px and CAT significantly (P<0.05); increased the contents of TG, TC, LDL-C, reduced the content of HDL-C in serum (P<0.05); decreased the activities of LPL and HL, increased the AI significantly (P<0.05). Compared with tea oil group and coconut oil group, lard oil diet and oxidated tea oil diet increased the level of FR in blood, decreased the T-AOC, the activities of SOD, GSH-Px and CAT significantly(P<0.05); increased the contents of TG, TC, LDL-C, decreased the content of HDL-C in serum(P<0.05);decreased the activities of LPL and HL, increased the AI significantly, decreased the mRNA expression levels of SS, CPT-1 and increase the mRNA expression level of ApoE in intestinal and liver. LA can reduce the bad effects to body caused by the oxidated tea oil diet and 0.1% is suitable.Conclusion: Oils which are different in saturation, length of carbon chain and oxidation degree have different effects on oxidative stress and blood lipid metabolism, while LA can reduce the degree of oxidative stress.
