Edible bird's nest(EBN)is a kind of nest made by male collocalia through its saliva and feather during mating period and has important edible value.Researches shows that the main nutrition in EBN are sialic acid,protein,reducing sugar and water,with little fat.Most of the studies on EBN are focus on the functions such as antivirus,anti-aging,anti-oxidation and improvement of bone strength by far.There are not much studies focus on the changes of the immunity of the female individuals and intelligence of pups during lactation caused by EBN.In this study,the female mice were treated with homogenate of EBN through intragastric administration during pregnancy,lactation and both pregnancy and lactation respectively,to analyze the content of the free and oligosaccharide-bounded sialic acid and protein-bounded sialic acid in mouse milk,milk yeilds were also measured after the birth of cubs in order to research the conditions of the pups' nutrition intake;the conditions of immunity of female mice were analyzed through the measurement of the spleen index,thymus index and the content of IL-2 in serum after the adult female mice were treated with EBN;the changes of intelligence of pups were analyzed through Morris Water Maze(MWM),the test of content of MDA,activity of SOD in cubs' brains and serum,the activity of ChAT and AchE in cub's palliums and hippocampi,theneuron count of hippocampi and the immunohistochemical staining of BDNF in cub's palliums and hippocampus.The results shows that:(1)the content of free sialic acid and oligosaccharide-bounded sialic acid and the milk yeild were significantly increased after the female mice treated with high-dose EBN in pregnancy and lactation period respectively or treated in both periods;(2)the most significant increase of immune organs index was seen in the group where female mice were treated with high-dose EBN homogenate in both pregnancy and lactation period;while intragastric administration with middle-dose EBN during lactation period or both pregnancy and lactation period can significantly rise the content of IL-2 in serum,the counterpart only during lactation shows non-significant effect;(3)in MWM test,pups in high-dose groups own significantly more times crossing the target area and longer time in third quadrant,compared with the cubs in middle/low dose groups;the groups treated with sialic acid solute have significantly more times crossing target area them the counterpart groups in different periods respectively,and except the groups treated with EBN homogenate through all the periods,longer time than the groups treated with low/middle-dose EBN group;the differences of intelligence among high-dose groups are not significant but still significant compared with the blank control group.(4)the activity of SOD in brian and serum andthe activity of ChAT in palliums and hippocampi of cubs in high-dose groups were relatively higher,with the lower content of MDA and activity of AchE;according to the photos,the amount of neurons and the expression of BDNF in cubs' palliums and hippocampi in high-dose group treated with homogenate during both pregnancy and lactation period were significantly higher than the ones in blank control group and the pregnancy&lactation sialic acid treated group;the fresh weight of brain and the total content of protein in brain show no significant difference among the three groups mentioned before.In summary,the intake of the high-dose of EBN during both pregnancy and lactation can increase the content of free sialic acid in milk and the milk yeild of female individuals.The female individuals who are fed with high-dose EBN during both pregnancy and lactation,have more significant increase in organ index after lactation.Combined with the level of IL-2,the female individuals in that group own the relatively better immune function.Besides,the female individuals fed with high-dose EBN during both pregnancy and lactation possess the filial generations lactated by them with better memory which related to the improvement of antioxidate ability and the development of neurons in hippocampi.
