Edible bird's nests(EBN)are nests made from saliva and feather by certan types of swiftlets(Apodidiae)in the breeding season.It's mainly produced in Southeast Asian countries like Indonesia,Thailand and Southwest area in China,and Austrilia.etc.The book "Compendium of Materia Medica" written by Li Shizhen records its function," bird's nest is good to kidney,stomach and spleen."It is very popular in the world since it has a high value of nutrition and medicine.At present,there are many studies about the functions of bird's nest,but few research on the effect of bird's nest on the recovery of specific damaged organs,and none research on the effect of bird's nest on the immune function to pneumonia animals.In this paper,the effect of EBN on immune function of pneumonia mice and brief mechanism.It will the basis of confirming the function of EBN to nourish and aftercare many lung injury diseases,and to provide experimental basis in the future to the further development of EBN.The results were determined as follows:1.The content of main nutritional ingredients of EBN homogenate was determined.The results showed that the content of total aminoacids,the protein,the sialic acid,fat,sugar and the ash are 49.49%(including 9 kinds of essential/semi-essential amino acid),59.53%,9.47%,0.04%,0.08%and 2.39%.EBN homogenate is rich in inorganic element,including macro elements(such as Ca,Mg,Na,K)and some trace elements.In a word,it has high value of nutrition.2.Using SO2 fumigation to found the model of pneumonia mice,and to study the effect of EBN homogenate of different concentrations on pneumonia mice's symptoms,weight,index of immune organs,pathological changes of lungs and the level of cellular factors(serum IL-2,IL-4 and IFN-y).The results showed that EBN homogenate of middle/high dose could improve the immune function of pneumonia mice to some degree.The EBN homogenate of middle/high dose the same as the Control group's mice didn't show the symptoms of coughing,tiring,poor appetite,poor luster,messy hair and hair-loss,irritability,significantly weight loss.The index of immune organs in the EBN homogenate of middle/high dose group,the same as the Positive Control group's mice,had no significantly changes compared with the Control group,but significantly-increased compared with the Model group.It indicated that mice in EBN homogenate of middle/high dosage group had slighter lung inflammation compared with Model group.The results of HE-staining of lung in mice showed that the inflammation of lung tissue in EBN homogenate of middle/high dose group's mice was between Model control group and Control group.The level of IL-2,IFN-y in EBN homogenate of middle/high dose group's mice were close to Control group,but significantly-decreased compared with the Model group.There was no significant difference between groups in the level of IL-4.It suggested that EBN might have a certain effect on the recovery of pneumonia mice,and the effect of the middle dosage group is better.3.Study on the effect of the EBN middle dosage on mice spleen lymphocyte proliferation and apoptosis of spleen lymphocytes in pneumonia mice,and the changes of IL-8,malondialdehyde(MDA)content and gene in pneumonia mice's lung tissue.The effect of the spleen lymphocyte proliferation of the EBN middle dosage group was significantly higher than the Control group(P<0.05),but significantly lower than the Model group(P<0.05),it showed that EBN homogenate may effectively alleviate the immune stress response on pneumonia mice's.The percentage of mice spleen lymphocyte early apoptotic cells and total apoptotic cells in model group was significantly higher than that of Control group(P<0.05).It showed that EBN can inhibit the apoptosis of splenic lymphocytes to a certain extent,because the percentage of early apoptotic cells in the EBN middle dosage group was significantly lower than that of Model group(P<0.05).The levels of IL-8 and MDA in lung tissue of mice in Model group were significantly higher than those of other groups(P<0.05),but there were no significantly difference in the levels of IL-8 and MDA between EBN middle dosage group and the Control group(P>0.05).It indicated that EBN may inhibit the inflammatory reaction and reduce lung injury in pneumonia mice through inhibiting the body to produce IL-8 and MDA.Compared with the Control group,there were three kind of common differential genes among the other three groups' lung:Atrophin-1,Cytokeratin-5 and Cyclooxygenase-2.EBN could inhibit the expression of Atrophin-1,Cyclooxygenase-2,delayed the injury of lung tissue in pneumonia mice,so as to enhance the body's immune function.
