Objective: To study the Preliminary safety evaluation of NCTF and hepatoprotective effects and its mechanism by D-Gal,Con A-,and CCl4-induced liver injury in mice on the basement of the safety evaluation,and further investigate its immunomodulatory effects simultaneously.Method: Preliminary safety evaluation of NCTF in mice by acute toxicity test.The hepatoprotective activities of NCTF(50,100 and 200mg/kg/day)was investigated by Con A(20mg/kg),D-Gal(800mg/kg)and CCl4 induced acute liver damages in mice.Dimethyl diphenyl bicarboxylate(DDB,200mg/kg)as a positive drug.Blood and liver were collected for biochemical studies and histopathological analysis.The preventive effect of NCTF on CCl4-liver damages in mice was investigated according to similar produre.The immune suppression model mice induced by cyclophosphamide,serum IL-4,IgM,IgG,serum hemolysin level(OD)and ACP,LDH content of spleen homogenate for detecting immune function indexes of NCTF.The phagocytosis of mouse peritoneal macrophages by experiment,carbon clearance index K index,further investigate its role in immune regulation.Result: The maximum tolerance dose of mice to MTD was 17 g/kg,suggesting that NCTF is safe and nontoxic to NCTF.Pre-treatment with nicotiflorin significantly(P<0.05)decreased Con A,D-Gal and CCl4 induced elevation in serum levels of ALT,AST as well as activities of pro-inflammatory cytokines IL-1?,IL-6,TNF-?,and INF-?.The hepatic anti-oxidant parameters(MDA,SOD,GSH-Px and NO)in mice with NCTF treatment were significantly antagonized for the pro-oxidant effects of Con A and D-Gal and CCl4 Histopathological studies also supported the protective effect of nicotiflorin.Compared with the model group,each dosage group of NCTF could improve the immune suppression of the hemolysin level(P<0.05),serum IL-4,IgG,IgM(P<0.05),and the level of mouse spleen homogenate ACP and LDH(P<0.05).3)compared with the model group,NCTF low,medium and high dose group(50,100,200mg/kg)could significantly increase the carbon clearance index of mice(P<0.05).Conclusion: NCTF has a significant protective effect on acute liver injury in mice induced by carbon tetrachloride and ConA D-galactosamine,induced acute liver injury in mice,induced by CCl4 in mice with chronic chemical liver injury,the mechanism might be related to anti-oxidant and decrease of releasing inflammatory cytokines.
