Objective: To evaluate the protective effect of extracts from Uighur medicine Anchusa italica Retz on myocardial ischemia reperfusion injury,and the results may be useful to expound the activity material base of medicinal herbs and lay the foundation fot the further development.Methods: 1)To establish a hypoxia/reoxygenation original generation of myocardial cells in mice in vitro model to simulate the ischemia-reperfusion injury.Experiment objestives were divided into 24 groups,normal control group,hypoxia/reoxygenation group,positive control group,(hypoxia/reoxygenation+1.25mg/mL puerarin),high,medium and low dose ethanol extracts treatment groups(hypoxia/reoxygenation+ high,medium and low concentration ethanol extracts from Anchusa italica Retz),high,medium and low doses of petroleum ether extract treatment groups(anoxia/reoxygenation+high,medium and low concentration petroleum ether extracts from Anchusa italica Retz),high,medium and low doses of ethyl acetate extract treatment groups(anoxia/reoxygenation+high,medium and low concentration ethyl acetate extracts from Anchusa italica Retz),high,medium and low doses of n-butyl alcohol extract treatment group(anoxia/reoxygenation+high,medium and low concentration n-butyl alcohol extracts from Anchusa italica Retz),high,medium and low dose water extract treated group(anoxia/reoxygenation+high,medium and low concentration water extracts from Anchusa italica Retz),high,medium and low dose the total flavonoids effective parts treated groups(anoxia/reoxygenation+high,medium and low concentration the total flavonoids effective parts of Anchusa italica Retz),High,medium and low dose rutin treatment groups(hypoxia / reoxygenation + high,medium and low concentration Rutin).MTT method was used to detect the survival rate of Primarycardiac myocytes,enzyme-linked immunoassay was used to detect the lactate dehydrogenase(LDH)levels in cell culture supernatant,malondialdehyde(MDA)content and superoxide dismutase(SOD)activity in cells.2)C57BL/6 mice ischemia reperfusion model is set up,and the C57 mice were divided into control group,the myocardial ischemia reperfusion group,high,medium and low doses of the total flavonoids effective parts of Anchusa italica Retz.The activity of serum myocardial enzyme CK-MB and heart markers CTn-I were determined by antigen antibody Elisa method,the myocardial infarction rate was determined by Evans blue TTC staining,and infarction zone of heart ischemia was stained with HE.Results:1)Compared with normal control group,a time-dependent decrease in primary myocardial cell viability in hypoxia/reoxygenation group was present.Compared with anoxia/reoxygenation group,treatment groups,the ethyl acetate extract,ethanol extract,n-butanol extract,water extract,total flavonoids effective parts,positive control and rutin significantly increased survival rate(P<0.01 or P<0.05),decreased LDH and MDA content(P<0.01orP<0.05),SOD activity significantly increased,the difference was statistically significant.According to evalution of time and concentration,the activity of ethyl acetate extract and n-butanol extract were stronger(P<0.01).2)Compared with the sham operation group,the percentage of myocardial infarction in the model group increased significantly,the difference was statistically significant(P<0.01).Compared with model group,the percentage of myocardial infarction in the groups of total flavonoid effective parts of Anchusa italica Retz significantly decreased,with statistical significance(P<0.01).Compared with control group,CK-MB and CTn-I levels in model group significantly increased,with statistical significance(P<0.01).Compared with model group,CK-MB and CTn-I levels in the groups of total flavonoid effective parts of Anchusa italica Retz decreased significantly,with statistical significance(P<0.01).Conclusion: 1)Rutin ethanol extracts,ethyl acetate extracts,n-butanol extracts and water extracts and total flavonoids from Anchusa italica Retz showed protective effect on myocardial ischemia reperfusion injury.The mechanism may be to reduce the oxidative stress induced by hypoxia / reoxygenation in primary myocardial cells by interfering with the production of oxidative stress products and enzyme system of antioxidant stress.2)Total flavonoid effective parts of Anchusa italica Retz has protective effect on myocardial ischemia reperfusion injury,and can reduce the rate of myocardial infarction and decrease the level of myocardial enzymes.
