Objective Analysis of 27 Y-chromosome STR markers in the population of Gansu Linxia Hui and Dongxiang ethnic minoritories and investigation of the genetic relationship of the male population of them,as well as evaluating their forensic application.Methods Under the organization of the Key Laboratory of the Department of Modern Anthropology of Fudan University and under the cooperation of local health administrative departments,following the principles of "informed consent",strictly in accordance with the sample access standards,a total of 301 Hui and 526 Dongxiang male venous blood(2 to 5 ml)were randomly collected in Gansu Linxia Hui Autonomous Prefecture.The 27 Y-STR loci were analyzed in unrelated healthy adult males from Hui(301)and Dongxiang(526)by STRtyper-27 Y kit(Health,China).The PCR products were analyzed by electrophoresis through 3130 XL genetic analyzer.The fragment sizes of alleles were analyzed subsequently by Gene Mapper ID v3.2.Allele/Haplotype frequencies and forensic parameters of 27 Y-STR were statistically analyzed and compared with the available data of other populations from different races and regions.The genetic relationship analysis was worked out with software Gen Al Ex6.502,SPSS20.0,Mega 5.0.Results In 301 Hui males,the gene(haplotype)diversity was from0.4668(DYS391)to 0.9686(DYS385a/b);295 haplotypes were observed,and the HD value was 0.9999,the discrimination capacity was 0.9801.In526 Dongxiang males,the gene(haplotype)diversity was from 0.4539(DYS391)to 0.9575(DYS385a/b);471 haplotypes were observed,and theHD value was 0.9973,the discrimination capacity was 0.8954.Population comparison reveals the genetic distance between Linxia Hui and Ningxia Hui was the lowest;genetic distance between Dongxiang and Xinjiang Uighur ethnic was the lowest.Conclusion The genetic distribution of 27 Y-STR loci in Gansu Hui,and Dongxiang ethnic minorities showed favorable polymorphisms,and was thus suitable for forensic identification and paternity testing in local area.The study of genetic diversity among different populations is useful for the research of their origins,migrations and their relationships.
