The Research On Quality Standard Of Periplaneta Americana

Objective:In this paper,we studied the quality standard of Periplaneta americana.The recovery from the processing of medicinal herbs,medicine identification and examination,determination of content of inquiry,to establish specific quality standard method of the optimum process conditions,the main quality components,improve and supplement local standards,to develop a more perfect Periplaneta americana reliable quality standard.Method:For Periplaneta americana and traits of harvesting and processing control,special inspection and record farming,harvesting,processing and storage process in the breeding base;with statistical characteristics and the observation of characters.TLC identification: the identification conditions of the local standard were compared,and the butanol ethanol water(2: 1: 1)was used as the developing agent,and the ethanol solution was used as the chromogenic agent,and the color of the solution was heated at 0.5% DEG C after heating at a temperature of 105 DEG C.Inspection items: by consulting a large number of relevant literature,combined with the local standards of Periplaneta americana and the inspection methods of the Chinese Pharmacopoeia,the inspection items of Periplaneta americana were studied,verified,supplemented and perfected.Determination of content: control content of medicinal materials,the total amino acid to alanine L-were determined by UV,and the detection wavelength was 570 nm.firstly optimized degreasing condition(degreasing degreasing time 1.5h,temperature 70 C solid-liquid ratio was 1:30),which can maximize the grease off medicine.Secondly,the orthogonal design method was used to optimize the extraction process of total amino acids from Periplaneta americana.The ethanol concentration was 80%,the extraction time was 1H,the extraction time was about 3 times,and the ratio of feed to liquid was 5: 75).Nucleotides with uracil,hypoxanthine and inosine components as control,were determined by HPLC method.The extraction conditions by single factor method first selected medicinal nucleotides composition: 65% ethanol,ultrasonic extraction method HPLC 1H;chromatographic conditions: column YMC-Park Pro-C18 column(250mm * 4.6mm,5 m).With 3% methanol(containing 0.07% acetic acid)and methanol as the mobile phase gradient elution;the detection wavelength was 250 nm,the column temperature was 25 degrees Celsius,the flow rate was 0.6ml/min.The methods of determination of amino acids and amino acids were studied.Results:The processing and processing of Periplaneta Americana is scientific,the yield is high,and it can provide the better quality raw material for the production.Through observation,description,statistics and comparative research,in length(27 ~ 35mm)and pronotum(with yellow and white background with a red brown butterfly shaped plaques)and local standards of Yunnan province and Hunan Province,local standards are different,the standard is more accurate and representative.The identification of thin layer,strong specificity,in the silica gel on the same position as the control can show more obvious red peach spots.The impurities shall not exceed 0.3%.Check on the limit of the moisture content shall not exceed 10%,2% higher than the local standard of 12%;the total ash not more than 6%,2% higher than the local standard of 8%;acid insoluble ash shall not exceed 0.5%,1.1% higher than the local standard 1.6%;extract the content of not less than 20%,and the local standard.Determination of content: the total amino acid content of not less than 3%,uracil concentrations of nucleotide components shall not be less than 250?g/g,not less than 650?g/g,inosine and hypoxanthine may not be less than 550?g/g.The contents of heavy metals in the 10 batches of Periplaneta americana were lower than twenty ppm,and the content of arsenic was less than one millionth of the total of two.Conclusion:This is the first study to study the quality standard of Periplaneta americana,were studied from the aspects of harvesting and processing,character,identification,examination,content,comparison of local standardgathers Periplaneta americana standard,improvement in the recovery process,and improve the standard characters and supplementary material,increase the basic quality indicators on behalf of Periplaneta Americana pharmacological and content control of TLC identification of TLC optimization results verify the superiority of the local standards of quality control.Check items,not of the ash content control limit increased soluble to water,total ash,acid,and heavy metals and arsenic salts to check,because the content of harmful to human value is far lower than the specified limit,the heavy metals and arsenic salts check items not included in this standard.This study was to establish a more practical,can reflect the better quality and efficacy of Periplaneta americana standards.
