Objective This study was designed to investigate the serum levels of inflammatory cytokines CCL-2,IL-18,TNF-α and TGF-β,and try to elucidate the correlation between the inflammatory factor levels and the cognitive function in patients with Alzheimer’s disease.Methods45 AD patients and 30 cognictively normal subjects whose age,gender is matched were recruited from the First Hospital of Jilin University from December 2015 to December 2016 in our study.Serum levels of CCL-2,IL-18,TNF-α and TGF-β were determined with enzyme-linked immunosorbent assay(ELISA).Mini-Mental State Examination(MMSE),Montreal Cognitive Assessment(Mo Ca),Memory and Executive Screening(MES),Clinical Dementia Rating Scale(CDR),Active of Daily Living scale(ADL)were used to assess the cognitive function and activities of day living of the subjects.SPSS software was used to analyze the results.ResultsThe serum levels of CCL-2、IL-18、TNF-α were significantly higher in AD patients than that of normal control(p < 0.05),while the serum levels of TGF-βwere significantly lower than that of normal control(p<0.05).The serum levels of CCL-2were correlated negatively with MES score(r=-0.243,p<0.05),and were correlated negatively with the write score of MMSE(r=-0.426,p<0.05)in AD patients.The serum levels of IL-18 were correlated negatively with the language tests score of Mo CA(r=-0.320,p < 0.05)in AD patients.The serum levels of TNF-α were correlated negatively with the write score of MMSE(r=-0.320,p < 0.05)in AD patients.The serum levels of TGF-β were correlated positively with MoCA score(r=0.276,p<0.05),and were correlated positively with the visuospatial abilities score(r=0.400,p<0.05),attention acore(r=0.274,p<0.05)and orientation score(r=0.279,p<0.05)of Mo CA in AD patients.The serum levels of TGF-β were also correlated positively with the orientation of time score(r=0.310,p < 0.05),attention and calculation score(r=0.279,p<0.05)of MMES in AD patients.Conclusion1.The peripheral serum levels of CCL-2、IL-18、TNF-α were significantly higher in AD patients than that of normal control.And the serum levels of TGF-βwere significantly lower than that of normal control.This suggests that serum levels of CCL-2,IL-18,TNF-α and TGF-βcould be a candidate marker for clinical diagnosis of AD.2.The serum levels of CCL-2 were correlated negatively with MES score.This suggests that the serum levels of CCL-2 could reflect the severity of AD.3.The serum levels of TGF-β were correlated positively with Mo CA score(r=0.276,p<0.05).This suggests that the serum levels of TGF-βcould reflect the severity of AD.
