Objective:This study is designed to investigate A1298 C and G1793 A polymorphism of MTHFR gene in Cerebral Infarction patients sampled in the population of Han nationality in Jilin Province.By comparing the genotyping and allele frequencies of A1298 C and G1793 A,possible correlation between the MTHFR gene polymorphism and the morbidity of cerebral infarction and the level of Hcy.Methods:In Jilin Province,336 subjects of Han nationality were studied and divided into 2 groups,control group and cerebral infarction group(CI group).There were 220 inpatients from Department of Neurology,the Second Hospital of Jilin University in CI group and 116 healthy people from Department of Neurology in control group.Collected the general situation of the study,history and related checks and other information.The genotyping of polymorphisms A1298 C and G1793 A of the MTHFR gene was performed by allele-specific polymerase chain reaction assay(AS-PCR).All data were processed by Excel 2003 and analyzed by SPSS 19.0.Results:1.The frequency of gene distribution in the cerebral infarction group and the control group was in accordance with the Hardy-Weinberg genetic equilibrium law.The two groups were genetically balanced(p>0.05).2.Compared with control group,subjects of CI group had a significant higher rate associated with hypertension?hyperlipidemia?gender?alcoholism and smoking(p <0.05).In terms of diabetes,there was no difference between the two groups(p>0.05).3.The levels of Hcy and LDL-C in the cerebral infarction group were significantly different from those in the control group(p<0.05).There was no difference in Uric acid level between two groups(p>0.05).4.In terms of genotyping of A1298 C polymorphisms,there was no statistic difference between two groups(?2=3.460,p=0.063).A allele frequency and C allele frequency in the cerebral infarction group and the control group between the distribution,the difference was statistically significant(?2=3.943,p=0.047).There was a significant difference between two groups in terms of genotyping of G1793 A polymorphisms(?2=14.328,p=0.001).Compared with control group,both AA genotype and A allele frequencies of G1793 A polymorphisms were significantly higher in the CI group(?2=13.438,p<0.001).5.The distribution of Hcy levels between A1298 C polymorphism AA and AC + CC genotypes was statistically significant(?2=7.702,p<0.001).G1793A polymorphism GG,GA,AA genotype between the distribution of Hcy levels,the difference was statistically significant(?2=92.942,p<0.001).6.According to logistic regression analysis,risk factors associated with cerebral infarction are age,gender,hypertension,LDL-C and G1793 A gene polymorphism.According to the OR: A1298 C gene polymorphism did not increase or reduce the risk of cerebral infarction;G/A heterozygous genotype did not increase or decrease the risk of cerebral infarction;The risk of cerebral infarction in AA genotype was 6.55 times higher than that in GG genotype population;Hyperlipidemia in the risk of cerebral infarction than people without hyperlipidemia increased by 1.51 times;The risk of cerebral infarction in hypertensive patients was 1.77 times higher than that in hypertensive patients;Compared with males,women are 0.507 times the male population.Conclusions:1.MTHFR gene A1298 C,G1793A genotype have polymorphism in Han nationality population in Jilin province.2.Elevated plasma levels of Hcy are associated with cerebral infarction.3.G1793 A polymorphism of MTHFR gene is related to the development of cerebral infarction in the Han nationality population in Jilin province.There is no correlative relationship between A1298 C polymorphism of MTHFR gene and the development of cerebral infarction in the Han nationality population in Jilin province.5.G1793 A allele AA homozygous mutations may be an important risk factor for the development of cerebral infarction in the Han population of Jilin province.
