Sarcodon imbricatus(l.e x Fr.)Karst is known as the national treasure in the history of our country.In this study,in this paper,the extraction ratio and bioactivity of polysaccharides which were extracted from fruit body of Sarcodon imbricatus(l.e x Fr.)Karst in Sichuan and Yunnan were studied and compared.Sarcodon imbricatus polysaccharides(SP)were extracted from fruit body of Sarcodon imbricatus(l.e x Fr.)Karst in Sichuan(SSP)and Yunnan(YSP).And the extraction process was optimized by response surface method.Building a model between the extraction ratio of water-soluble polysaccharides and these dependent factors.The optimum extraction conditions were confirmed as follows: YSP: 3.1 h,60:1,91?,and the highest extraction rate of polysaccharides was 16.75 %;SSP: 3.1 h,58:1,93?,13.93 %,which was significantly higher than the previous maximum extraction rate of 11 %.In this antibacterial experiment,the ethyl acetate extract,the n-butanol extraction phase and the polysaccharide phase were selected as the samples.The antibacterial activities of of each phase was determined for Aspergillus niger,Pseudomonas aeruginosa,Escherichia coli,Staphylococcus aureus,Bacillus subtilis and Bacillus subtilis by inhibition zone test.The results showed that ethyl acetate extract had some antibacterial activities for Bacillus subtilis,Escherichia coli and Staphylococcus aureus,and the bacteriostatic effect of Bacillus subtilis was the highest significant(inhibition zone diameter> 12 mm)from Sichuan;The ethyl acetate extract from Yunnan had antibacterial activity for Bronchobacterium,Escherichia coli and Bacillus subtilis,Which had the most significant bacteriostatic effect(the inhibition zone diameter> 14 mm).In this study,the YSP and SSP were used as the experimental samples,Vc as the positive control.Furthermore,the antioxidant activities of polysaccharides were investigated in three aspects,namely 1,1-diphenyl-2-picrylhydrazyl(DPPH)radical,hydroxyl radical and super-oxide anion scavenging activity.The results showed that the antioxidant activities of SP was dependent on their concentrations.And the highest scavenging rates of SSP to DPPH,superoxide anion and hydroxyl radicas were 86.14%,71.78% and 99.98%,respectively,which were higher than 76.54 %,58.52 %and 99.93 % of YSP,and their hydroxyl radicals clearance was effective than 87.97 % of Vc.What's more,the results showed that the antioxidant activity of SSP-1(enzyme extraction)was higher than that of SSP(water extraction).Based on the results of anti-oxidative activities of SP in vitro.In this study,the anti-oxidative and the immunomodulatory activity in vivo of SSP in immunosuppressed mice were studied.The levels of MDA,SOD,GSH-Px and T-AOC in serum and liver homogenate were determined by immunosuppressive model and astragalus polysaccharide(AP)as positive control.At the same time,the immune activity of SSP was evaluated by immune organ index,delayed postharvest allergy(DTH),immunoglobulin and IL-2 production level and oxidative factor level in mice.The results showed that SSP enhances immunity of CTX-induced immunosuppressed mice via increasing the serum of immunoglobulin and immune factors,enhancing organ indexes of thymus and spleen,and neutralizing oxidative stress.
