Objective:By observing the atherosclerosis model in rats liver and abdominal aortic pathology change,Observation on the liver organization soft lecithin cholesterol acyltransferase(LCAT),ATP binding box transporter A1(ABCA1)expression and the effect of discuss sappanwood ethyl acetate extract of atherosclerosis in rats the effect of reverse cholesterol transport,as well as wood ethyl acetate extract of the mechanism of action of prevention and treatment of atherosclerosis and provide experinjental basis for clinical application of wood.Methods:By repeated injections of vitamin D370 all IU(kg/d)and a high-fat diet,the method of SD rats induced atherosclerosis model was established.40 adult rats,randomly divided into 4 groups and blank control group,model control group,respectively,the ethyl acetate extract of sappanwood group,simvastatin group,each group of 10,adaptability to feed a week later,in addition to more than the blank control group were intraperitoneal injections of vitamin D370 IU(kg/d)for 3 days,at the same time,fed a high-fat diet for 12 weeks,atherosclerosis model was established.After the success of the model to copy each dosing 28 days in a row,materials.Using HE staining to observe the change of the abdominal aorta and liver pathological morphology,RealTime PCR method is used to observe the liver LCATmRNA and ABCA1mRNA expression quantity.Results:? Then liver tissue:blank control group did not see abnormal liver tissue structure,centered around the central vein radiated liver cells,liver cells of structure is clear,no fatty degeneration of the cells;Model group rats hepatic cell swelling,accompanied by a large number of redundant adipose droplets;Sappanwood acetic ester group in the rat liver mild edema,did not see fat sample cavity formation;Simvastatin group rat hepatocyte swelling,accompanied by sample degeneration,tiny balloon sample duality can be changed.? In blank control group within the abdominal aorta vascular wall,and a clear demarcation between the outer membrane;Model control group under the intimal smooth muscle cells are arranged disorder,hyperplasia,visible tiny foam sample cells appeared at the same time,the elastic fiber layer degeneration;Sappanwood acetic ester group of endothelial cell surface is smooth,a small amount of disordered arrangement of smooth muscle cells and hyperplasia,the membrane elastic fiber layer is irregular,outer membrane for thin loose connective tissue;Simvastatin group lining under visible with a small amount of inflammatory cells and lipid deposition and smooth muscle cell proliferation and disordered arrangement,and have arranged irregular,membrane elastic fiber layer in the outer membrane thinning.?RT-PCR results showed that compared with the blank group,each group LCATmRNA expression in liver tissue significantly reduced,statistically significant difference(P<0.05);Compared with model group,the ethyl acetate extract of sappanwood group and simvastatin group significantly increased the expression of liver tissue LCATmRNA,statistically significant difference(P<0.05);Ethyl acetate extract of sappanwood group with simvastatin group compared the liver tissue LCATmRNA expression difference no statistical significance(P>0.05).?RT-PCR results show that compared with the blank group,each group ABCA1mRNA expression in liver tissue decreased significantly,the difference is statistically significant(P<0.05);Compared with model group,the ethyl acetate extract of sappanwood group and simvastatin group significantly increased the expression quantity of ABCA1mRNA in liver,statistically significant difference(P<0.05);Sappanwood acetic ether extract group and simvastatin group there was no statistically significant difference comparing ABCAlmRNA expression in liver(P>0.05).Conclusion:1.The Sappanwood acetic can alleviate the pathological damage of liver and abdominal aorta in rats with atherosclerosis.2.The Sappanwood acetic could improve expression of LCATmRNA and ABCA1mRNA in rats with atherosclerosis.
