| Objective:To observe the effect of Jianpi Yiqi Formula on the pathological changes of gastric mucosa in CAG rats established by MNNG combined with 0.03%rarnitidine,to observe the expressions of GAS,PG E2,PP,and ?-catenin,Cyclin D1,GSK-3p,et al,for explore the possible mechanism of Jianpi Yiqi Formula.Methods:The model rats are established by 150?g/mL MNNG combined with 0.03%ranitidine pelletized feed for 24 weeks.Forty rats are divided into 4 groups averagely:normal group(normal feeding),model group(0.9%Nacl),positive group(3.16mg/kg/d folic acid+15.80 mg/kg/d teprenone)and Jianpi Yiqi Formula group(14.04g/kg/d).Except for the normal group of normal feeding,the rest rats are gavaged for next 8 weeks,then all rats are killed,and serum and gastric tissue samples are collected.Observe the general situation of the rats,serum levels of GAS and PG E2 are measured by ELISA,tissue PP are measured by colorimetric detection,and observation of pathological morphology of gastric mucosa by H&E Staining,the expression of GSK-3?,?-catenin and Cyclin Dl protein in gastric tissue are observed by imunohistochemistry.Results:?The weight of rats:The weight of the normal group,the positive group and the Jianpi Yiqi Formula group are increased significantly(P<0.01),the model group are not obvious(P>0.05);and Positive group and Jianpi Yiqi Formula group are significantly higher than the model group(P<0.01),but there is no significant difference between these two groups(P>0.05).?Pathomorphological changes:Normal group of rats have gastric mucosal shape of the rules,arrange closely and neatly;In the model group,the gastric mucosa thinning,the glands reduce,the gastric pits and the propria thick;The mucosa of the positive group is rare or occasionally atrophic,and the degree of mucosal structure disorder is lighter than that of the model group;Jianpi Yiqi Formula group of rats gastric mucosal structure is clear,rare atrophy,lamina propria thickening.?Levers of serum GAS,PG E2 and tissue PP:The levels of GAS,PG E2 and PP in the model group are significantly lower than those in the normal group,and in positive group and Jianpi Yiqi Formula group are higher than those in model group(P<0.05 or<0.01),there is no difference between these two groups,but Jianpi Yiqi Formula group shows the advantages(P>0.05).?Expression of GSK-3?,?-catenin and Cyclin Dl:The mean optic density(MOD)of GSK-3(3 positive cells in the model group is lower than normal group(P<0.05),Positive group is slightly higher than the model group,but no significant difference(P>0.05),the MOD of Jianpi Yiqi Formula group is higher than the model group(P<0.05).The MOD of(3-catenin positive cells in the model group is higher significantly than that in the normal group(P<0.01),The MOD of Positive group and Jianpi Yiqi Formula group positive cells is lower than the model group(P<0.05).The MOD of Cyclin D1 positive cells in the model group is higher significantly than normal group(P<0.01),Positive group and Jianpi Yiqi Formula group positive cells is lower than the model group(P<0.05 or<0.01).There is no difference between these two groups,but Jianpi Yiqi Formula group shows the advantages(P>0.05).Conclusions:?Jianpi Yiqi Formula group can improve the pathological state of gastric mucosal atrophy in CAG rats,regulate the expression of GAS,PG E2 and PP and GSK-3?,P-catenin and Cyclin D1.?We analyze that Jianpi Yiqi Formula plays therapeutic effectsare related to improve the lever of GAS,PG E2 and PP to improving the gastric mucosal acid function,enhancing the mechanism of gastric mucosal barrier;?Jianpi Yiqi Formula is related to regulate the expressions of Wnt/?-catenin signaling pathway related proteins GSK-3?,?-catenin and Cyclin D1 to regulating gastric mucosal cell differentiation,proliferation.In this study,we reveal some molecular mechanisms of the effect of Jianpi Yiqi Formula on CAG,and provide the basis for the study of the mechanism of the effect of Jianpi Yiqi Formula on CAG. |
