Experimental Research On Cigu Guabei Formula In Preventing The Postoperative Recurrence And Metastasis Of Breast Cancer

Background:Breast cancer now is the most common malignancy in women,and has seriously affected women's physical and mental health,even life-threatening.At present,the methods of treating breast cancer are surgery,radiotherapy,chemotherapy,endocrine therapy,molecular targeted therapy,Chinese medicine treatment and so on.This research will take the CiGu GuaBei Formula as the research object based on the clinical experience of Professor Li Yongjian for many years,which has significant effect in treating breast cancer.So,we will study the anti-cancer effect and the mechanism of it.Purpose and significance:In this research,we establish the model of mouse orthotopic breast cancer to observe the effect of CiGu GuaBei formula on the formation,growth,pulmonary metastasis of breast cancer in mice,as well as the effect of the survival and immune function of tumor-bearing mice.Meanwhile,it will provid experimental basis and foundation for the clinical research on CiGu GuaBei formula in preventing the postoperative recurrence and metastasis of breast Cancer.Methods:We select 50 healthy BALB/C mice and establish the model of mouse orthotopic breast cancer by injecting the mouse breast cancer cell line 4T1.At the same time,the model of mouse orthotopic breast cancer is divided into 5 groups according to the dosing:chinese medicine group(high concentration group 2.4g crude drug/mL,medium concentration group 1.2g crude drug/mL,low concentration group 0.6g crude drug/mL),blank group and positive control group(adriamycin).By comparing the time of tumor formation,the effect of drug intervention on tumor formation can be observed.To survey the drug intervention on tumor growth,tumor volume and tumor weight changes are measured respectively.In order to observe the effect of drug intervention on tumor metastasis,we stain the lung sections of tumor-bearing mice and measure serum TGF-?1 The analysis of whether the survival time of mice will be extended after drug intervention by comparing the length of life.The effects of drug intervention on immune function are observed by calculating the spleen index and thymus index of tumor-bearing mice.Results:The survival time of the mice in chinese medicine group is higher than that of other tumor-bearing groups,which proved that the CiGu GuaBei formula has the effect of prolonging the survival time.Compared with the blank group,the tumor volume and tumor weight of chinese medicine group are much smaller and lighter.It is proved that the CiGu GuaBei formula can inhibit the growth of 4T1 breast cancer cells in a certain extent,which is statistically significant.During the course of the operation,the lobes of the thymus in saline group are unclear and its volume is reduced,and the color of it is gray.Also,the color of the spleen is lighter than that of the chinese medicine group.However,in chinese medicine group,the color of the spleen is deep red and its volume is swelling.The results show that the CiGu GuaBei formula can motivate effective immune function to play a role in inhibiting the tumor by stimulating the immune organ hyperplasia in the process of treating the tumor.The spleen index and thymus index of chinese medicine group are significantly increased,indicating that the CiGu GuaBei formula has a protective effect on the immune organ.By observing the effect of drug intervention on tumor metastasis,it is find that the number of pulmonary nodules is significantly lower than blank group,and serum TGF-?1 in traditional Chinese medicine group has obvious increasing trend compared to blank group,which proved that the CiGu GuaBei formula can effectively inhibit tumor metastasis.Conclusions:In this study,we take the CiGu GuaBei formula as research object,observing that the CiGu GuaBei formula can inhibit the growth of breast cancer cells and prolong survival in a certain extent.Also.it can motivate effective immune function to play a role in inhibiting the tumor growth by stimulating the immune organ hyperplasia.At the same time,it can effectively inhibit tumor metastasis.
