Yangjing Capsule Improving The Microcirculation Of The Testis By Sertoli Cells Can Promote Mechanism Of Spermatogenic Function

Objective:Observation of Yangjing capsule extract on Sertoli cells of the reproductive ability of mice,and the effects of YCE on the Occludin,VEGF and its receptor,NO and its synthaseMethod:The mouse Sertoli cell line TM4 was used as the target of the experiment,and the cell proliferation activity was measured by the CCK-8 method when the cells were cultured for 70%hours,and the corresponding concentration of YCE was continued for a period of up to 12 hours.In the growth of TM4 cells in the logarithmic phase to collect the digestive cells,adjust the concentration,adding 6 hole plate of each hole about 80 thousand cells with different concentrations of YCE to continue after 24 hours of incubation,supernatant directly to the determination of the NO content,a collection of cells,from which the mRNA or protein,respectively using qPCR and Western-Blot to analyze the Occludin and VEGFA,FLT and FLK,eNOS(NOS3)expression in.Results:The effect of YCE on TM4 cell after 12 h,and the proliferation activity of TM4 cells was significantly higher than that of the control group,which could enhance the activity.After 24 hours of YCE on TM4 cells,found no significant difference between VEGFA and mRNA protein compared with control group(P>0.05);and YCE of Occludin,VEGFA and FLK in each dose group compared with the control group at the mRNA and protein expression were increased Significantly(P<0.05);the effects of YCE on the FLT and eNOS when compared with the control group,there was no significant difference between the low dose group,Moderate and high dose group,significant difference(P<0.05).Conclusion:YCE could enhance the proliferation ability of TM4 cells,the expression of mRNA and protein in Occludin,VEGF receptor and NO synthase in TM4 cells and the content of NO,and there was dose dependence between them in YCE.It proves that Yangjing capsule can improve the proliferation ability of TM4 cells,enhance the spermatogenic environment,increase VEGF and NO content increase in testicular microcirculation,enhance local metabolism,improve the spermatogenic environment.
