| Objective:Qingfei Oral Liquid is effective for viral pneumonia in children,but its mechanism is still not so clear.In order to further demonstrate this mechanism,this study investigated the effects of Qingfei Oral Liquid on the expression of Interleukin(IL)-2,Interleukin(IL)-17 and the balance of Treg/Thl 7 in mice’s lung tissue infected with respiratory syncytial virus(RSV).By observing the changes of this index,we can elicit the mechanism preliminarily.Method:The research was conducted by dividing the fifty BABL/c mice into five groups randomly,namely,control group,model group,Ribavirin group,Qingfei Oral Liquid low dose group(1.33g·mL-1)and high dose group(4g·mL-1).ten mice per group.In addition to the control group,Other groups were molded by RSV(long strain)virus and given 48-hours’ cultivation.24 hours later they were killed and take the lung tissue for pathological analysis immediately.We can observe the status of alveolar wall edema,lung congestion,infiltration of inflammatory cells.ELISA test of expression levels of IL-2 and IL-17 in BALF and flow cytometry test of the level of Treg and Th17 in the spleen.We can find the mechanism by observing the changes of this index.Result:Pathological analysis showed that.the lung tissue pathological score of model group increased obviously compared with the control group.Compared with the model group,the lung tissue pathological score of Ribavirin group,Qingfei Oral Liquid high dose group decreased(P<0.05).Detection of Treg/Thl7 and related cytokines showed that.Compared with the control group,the level of IL-2 and Treg/Thl7 decreased and the level of IL-17 increased in the model group(P<0.01);Compared with the model group,both Qingfei Oral Liquid groups and Ribavirin group showed a significant increase of expressions of IL-2(P<0.01)and Treg/Th17(P<0.05)showed a decrease of expression of IL-17(P<0.01);Compared with the Ribavirin group,the Qingfei Oral Liquid groups showed a significant increase of expressions of IL-2(P<0.01).Qingfei Oral Liquid high dose group showed a increase of expression of Treg/Th17(P<0.05)and decrease of expressions of IL-17(P<0.01).Qingfei Oral Liquid low dose group showed a decrease of expression of Treg/Thl7(P<0.05)and increase the expressions of IL-17(P<0.01).Conclusion:1.Qingfei Oral Liquid can regulate the balance of Treg/Th17,increase the expressions of IL-2 and decrease the expressions of IL-17 in the lung tissue of RSV infected mice,through which the Qingfei Oral Liquid can play a role of anti-RSV.2.The experimental results show that Qingfei oral liquid for the treatment of RSV pneumonia is dose-dependent,with high doses more effective.Appropriate dose increase,its treatment of RSV pneumonia is superior to Ribavirin group.3.Traditional Chinese medicine has a good prospect in the treatment of RSV pneumonia.This study not only confirmed the effectiveness of Qingfei oral liquid for the treatment of RSV pneumonia,but also provides a way to investigate its pathogenesis. |
