Objective: Study on artificial cultivation of Dendrobium candidum water extract,on the influence to the streptozotocin(STZ)induced hyperglycemic mice's blood glucose,blood lipids and liver antioxidant levels,and the repair function to the vivo intestinal flora diversity.Through a variety of ways to analysis the damage of diabetes on the body,discuss the protective effect on the body and the possible mechanism.Methods: To establish STZ diabetic mouse model,randomly seperated to high,medium and low dose dendrobium extract group,normal control group,model control group and positive drug group.Observethe experimental animals' general situation,test the mice's body weight,determination of body composition in mice by NMR,with blood glucose test strips to fasting test blood glucose,serum TC,TG,HDL value detection in accordance with the kit,using the spectrophotometer to measure the content of liver SOD(xanthine oxidase method),measuring the content of MDA(TBA),through PCR-DGGE andquantity one software to analysis of intestinal bacteria diversity of mice.Results:(1)The mice in the normal control group were in good growth condition and no death.The model mice acted flagging,slow response and less activities,the amount of daily drinking and frequency of urination increased,and thebody weight decreased significantly(P<0.05),thehair became loose and the spirit became dull,irritability,the faeces became small,yellow and dry,the survival state of the model group was the worst,and the survival rate was only 50%.After each dendrobium dose group and positive medicine gavage treatment,the general state of dendrobium in each dose group and positive drug group's mice were better,the colour and the mental state had been restored,the quantity of water intake and urine decreased,and the faeces became-normal,the survival rates were 80%,60%,60%,and 80%,respectively.In the normal control group,with the increase of body weight,the adipose tissue was significantly increased,which was higher than that of other groups.In the model control group,with the decrease of body weight,compared with other groups,the adipose tissue was significantly reduced.Thedendrobium water extract of three dose groups and positive drug group,consistent changes in both adipose tissue and body weight,whether the proportion of fat or lean was higher than that of model control group,and the lean ratio was higher than the normal control group.(2)Compared with the normal control group,the model control group's blood glucose level increased significantly,which began to decline on 28 d,each dose of dendrobium water extract groups' blood glucose level began to decrease significantly after 7d(P<0.01),during 14d~28d,the blood glucose level were significantly decreased(P<0.01);Three dose groups of dendrobium water extract's function closedto positive drug group in the hypoglycemic effect,no significant difference between each dose(P>0.05).(3)After 28 days of experimental,compared with normal control group,the level of TC and TG in serum of model control groupwere signifcant higher(P<0.05),the level of HDL wassignifcant lower(P<0.05).Compared with the model group,the serum of TC content in three dose groups of dendrobium water extract and positive drug group decreased significantly(P<0.05),the content of HDL increased significantly(P<0.05).Both low dose group and positive drug group's TG level was lower(P<0.05),TG content of middle and high dose dendrobium group was higher than that of the model control group(P<0.05).(4)Compared with the normal control group,the SOD activity of liver tissue in model conrol group was significantly lower(P<0.05),the MDA level was significantly increased(P<0.01).Compared with model control group,three dose dendrobium water extract groups' SOD activity in the liver tissue were significantly increased(P<0.05),MDA content decreased significantly(P<0.05);the positive drug group's SOD activity increased,there was significant difference(P<0.05),but the content of MDA was higher than the model group.(5)Compared with the normal control group,the bacteria bands of positive drug group and model control group were blurred,showed low expression level.The water extract of dendrobium in the high dose group showed high bacterial diversity and expression.The results of cluster analysis showed that the water extract of dendrobium in low and medium dose group as a cluster of bacteria,had the highest similarity,the water extract of dendrobium in high dose group,model control group,positive drug group and normal control group as a cluster of bacteria,had the highest similarity.The positive drug group and normal control group of bacteria highly similar,in the water extract of dendrobium treatment groups,the high dose group and normal control group were most similar.To sum up,the middle and high dose group can improve the number and diversity of intestinal flora in diabetic mice.Conclusion:(1)Dendrobium water extract can improve diabetic mice body health and rapid weight loss disease;(2)Dendrobium water extract has a significant hypoglycemic effect in STZ-induced diabetic mice,and equivalently to the positive drug(Metformin Hydrochloride);(3)Dendrobium water extract can reduce TC and TG content that diabetes leads to increased,adjust the content of HDL,improve the dyslipidemia in diabetic mice;(4)Dendrobium water extract can adjust the diabetic mice's the antioxidant ability of liver,increase the activity of SOD,reduce the level of MDA;(5)Dendrobium water extract can regulate disordered intestinal flora,increase the number of bacteria caused by diabetes.In summary,the water extract of dendrobium can regulatethe level of blood lipid,antioxidant capacity of liver and the diversity of intestinal microflora,through improving lipid metabolism in diabetic mice,enhancing the immune ability,to indirectly regulate the stability of blood glucose level.So,the dendrobium can be used as adjunctive therapy in the treatment of diabetes,and can reduce the level of blood glucose and improve complications due to diabetes,provide new ideas for the prevention and treatment of diabetes in the future.
