With the increase of economy,people's diet habit changed,alcohol consumption is increasing,but heavy alcohol consumption can cause liver damage,which will further induce alcoholic liver disease(ALD).However,the treatment of ALD is lacking of efficient therapy.Therefore,looking for a safe and efficient treatment is necessary.Taurine,as a natural free amino acid,exists abundant in animals and has various physical functions.This study aims to explore the protective effects of taurine on ethanol-induced hepatocytes injury in vitro by detecting the Cell viability and apoptosis.180-220 g male Wistarrats were used to separate the original liver cells by Seglen two-step perfusion in situ combined by mechanical digestion method.The cells were identified by glycogen staining and the 3rd-5th generation were used for the following experiment.The experiment were divided into 5 groups:normal control group(C),model group(M)with 75 mmol/L ethanol in the medium,taurine treatment group(M+T)with 75 mmol/L ethanol and 10 mmol/L taurine in the medium,taurine control group(T)with 10 mmol/L taurine in the medium.48h later,inhibition rate of the cells were detected by MTT,concentrations of ALT,AST and GGT in the medium were detected.AO-EB staining were used to detect the apoptosis of hepatic cells.Total RNA and protein were extracted,Real-Time PCR and Westerning-blotting to detect the expressions of the apoptosis regulating factors.The results showed that 75mmol/L ethanol in vitro could damage 50%hepatocytes,which indicating the success of model establishment.10mmol/L taurine could significantly alleviate the inhibition rate induced by ethanol.The concentrations of AST,ALT and GGT in the model group increased significantly compared with the normal control group and the taurine treatment group,indicating that taurine could reduce the injury caused by ethanol.Immunocytochemistry staining found that apoptosis in the model group was obviously higher than the normal control and taurine treatment groups.mRNA and protein expressions of Fas,FasL,Caspase-3,Caspase-9 and Bax increased significantly,while mRNA and protein expressions of Bcl-2 decreased significantly in the model group compared with the normal control and taurine treatment groups.The results indicated that taurine exerts protective effects on the liver cells injured by ethanol,the mechanism of which may through that taurine could protect the membrane of the liver cells,reduce the fat deposition,and regulate the intrinsic and extrinsic apoptosis pathway to reduce the abnormal apoptosis induced by ethanol.
