Objective:Through the observation of ZhuangTongYin(ZTY)on myocardial ischemia of coronary heart disease and blood stasis syndrome of Ang II,TXA2 and PGI2,to explore the mechanism and therapeutic effects of the prescription on blood stasis syndrome of coronary heart disease myocardial ischemia,and provide basic information for the treatment of coronary heart disease myocardial ischemia and blood stasis through ZTY.Meanwhile,it can provide the research ideas and strategies for the development of the Guangxi characteristic Zhuang Medicine to prevent and cure coronary heart disease.Methods:Coronary heart disease myocardial ischemia blood stasis syndrome was induced by ligation of left coronary artery in rats.All rats were randomly divided into the following 7 groups:Blank group,sham operation group,model group,low,middle and high dose group in ZTY,and positive control group.The treatment groups were given intragastric administration every day.The ZTY groups were treated with ZTY(with 6.8g/kg for the ZTY low dose group,13.6g/kg for the ZTY medium dose group,27.2 for the ZTY high dose group).And given 80mg/kg Compound Danshen dripping pill as the positive control group.In the process of modeling experiment observed the change of electrocardiogram(ecg)in rats.After 4 weeks of experimental treatment,general situation and the dynamic change of electrocardiogram(ecg)of rats at each group were observed,the expression of AngII,TXA2 and PGI2 was detected by enzyme linked immunosorbent assay(ELISA)and real-time fluorescence quantitative RT-PCR.Numerical value was presented with the mean±standard deviation((?)±s).Data were analyzed using one-way analysis of variance(ANOVA).The criterion for statistical significance was set if P value was less than 0.05.Results:The general situation of model group is that they appeared gradually to be dry-yellow hair,squamous and dry skin,frightened easily,bend over,purple tongue and so on,and abnormal Q wave in ECG I,II,avL lead.Compared with the model group,the high,middle and low dose group and the control group had different degrees of improvement.Compared with sham operation group,AngⅡand TXA2 in model group and treatment group increased(P<0.05),PGI2 reduced(P<0.05).Compared with the model group,the AngII and TXA2 of rats in each treatment group were significantly reduced(P<0.05),PGI2 increased(P<0.05).Compared with the control group,there was no significant difference in AngII,TXA2 and PGI2 of the rats in each dose group(P>0.05).Conclusion(s):(1)The examination results above suggest that ZTY has certain curative effect on Coronary disease through reducing degree of heart Blood stasis,slowing progress in Coronary disease.(2)The examination results above suggest that ZTY has certain curative effect on myocardial ischemia and blood stasis syndrome of coronary heart disease through decrease the level of AngII in order to inhibit the activation of RAAS system in rats,decrease TXA2and the increase of PGI2 level. |
