Objective: Thin layer chromatography(TLC)was used to research The identification and study of the active constituents of Euonymus fortunei in guangxi.Response Surface for dulcite of Euonymus fortunei optimized extraction technology and determination of content of research.Euonymus fortunei the medicinal materialspicked the different regions of guangxi and studied by means of IR fingerprint and HPLC fingerprint.Doing the things to better provide a scientific basis for the development and utilization of the guangxi national medicinal materials for The quality control of Euonymus fortunei.Methods: 1.The extraction method was carried out by TLC method.The extraction method was carried out with 30% ethanol ultrasonic extraction method.The silica gel G plate was used as the thin plate the sample volume was 4ul the pyridine-ethyl acetate-Water-glacial acetic acid(8:15: 4: 1)as a developing agent.2.Infrared spectroscopy(FTIR)were used to identify Euonymus fortunei and the infrared fingerprints and herbs shared patterns were established.SPSS 22 statistical software was used to identify and correlate 25 batches of samples Evaluation and cluster analysis.3.The method of response surface methodology was used to optimize the extraction process.The central value was determined by single factor examination of liquid chromatography conditions and extraction methods.According to the design principle of Box-Benhnken’s central combination test the experimental results were analyzed by Design-Expert v8.06 statistical software for Response surface regression analysis.And then testing the methodological investigation and content determination.4.The method of preparation and the method of liquid chromatography were used to study the HPLC fingerprints of the medicinal materials and the common pattern of the medicinal materials was established.The traditional Chinese medicine developed by the National Pharmacopoeia Commission was established.Chromatographic Fingerprint Similarity Evaluation System 2012.1 Edition software for 10 batches of samples were identified with the common peak and similarity evaluation.Clustering analysis and principal component analysis of HPLC fingerprints were performed by SPSS 22 statistical software.Results: 1.Thin layer chromatography and infrared spectroscopy were all discriminative.And the common fingerprints of the infrared fingerprints were 8 and the similarity of the fingerprints of 25 samples was more than0.9.Through cluster analysis 25 batches of samples from different areas were divided into five groups.2.The extraction process was optimized by response surface methodology based on HPLC-ELSD method.The optimum extraction process was carried out with 32% ethanol 1:10material and 65 min extraction time.Under this condition The extraction rate of spear alcohol is 2.915%(Y)of the peak area and the natural logarithm(X)of the injection volume showed a linear relationship with the HPLC-ELSD content method.The sample injection rate is in the range of 2.24 ug ~ 22.4ug the sample precision and stability are good the recovery rate is 99.47%.3.A total of eleven peaks were identified by chromatographic evaluation system software.Eight peaks with good stability strong absorption peak and obvious peaks were selected as common peaks With the reference substance identified one of the quercetin total peak.The similarity of fingerprints of 10 batches of Euonymus fortunei was similar to that of 0.9 and the other was more than0.9.By cluster analysis 10 batches of samples from different areas were divided into two groups and the cumulative variance contribution rate of the three principal components was 90.26%.Conclusion: The method of thin layer chromatography on the identification of Phoebe burgdorferi in Guangxi was established.The characteristics of these infrared spectroscopy could be used to identify the main components contained in the medicinal materials accurately quickly and effectively.The extraction technology and content determination of the phytophthora from the different areas of Guangxi were determined and the HPLC fingerprints were established which provided the basis and theoretical reference for the quality evaluation of Euonymus fortunei in Guangxi.
