Aim: to explore the expression of PLCE1 and its value for predicting prognosis in hepatocellular carcinomaMaterials and methods:(1)The paraffin blocks of 90 cases of hepatocellular carcinoma confirmed by postoperative pathology as well as clinicopathological and prognosis data were collected.Immunohistochemistry was applied to determine the expression of PLCE1 in HCC and corresponding surrounding tissues from these section samples.Statistical analyses were used to examine the association between PLCE1 levels and clinicopathological features as well as prognosis.(2)PLCE1 levels in 20 paired HCC tissues and corresponding paracarcinomatous tissues was investigated by quantitative RT-PCR(polymerase chain reaction)and Western blot assays.(3)In addition,protein levels of PLCE1 in one normal liver epithelial cells and 4 HCC cell line were examined using Western blot assay.Results:(1)immunohistochemical(IHC)assay showed PLCE1 expression was significantly low in HCC tissues compared to the adjacent normal liver tissues(40% vs.18.9%;P<0.05).Besides,PLCE1 levels was negatively correlated with tumor capsulae,vascular invasion,Edmondson grade,AFP and TNM stage.Univariate analysis revealed that lower level expression of PLCE1 was significantly associated with poorer overall survival(OS)rate(P<0.001)and disease-free survival(DFS)rate(P<0.001).Multivariate analysis revealed low PLCE1 level was an independent poor prognostic factor of OS and RFS(P<0.001 and P=0.003,respectively).(2)qRT-PCR and western blot assay.qRT-PCR assay revealed PLCE1 levels were significantly lower in tumor sample tissues than that in adjacent nontumor tissues(0.51±0.49 VS.1.46±1.10,P = 0.001),and 85% of samples(17 paired of 20)were shown tumor tissues have a lower expression.(3)Western blot assay suggested PLCE1 levels in tumor sample tissues and adjacent paracarcinomatous tissues samples were 0.93±0.39 and 1.22±0.40,respectively(P=0.026),and 18 paired of these samples(18 of 20)were remarkable lower in HCC tumor samples,which showed a similar pattern to qRT-PCR assay.In addition,Western blot assay found PLCE1 was downregulated in all 4 HCC cell lines compared to the normal normal liver cells(THLE3).Conclusion: In brief,our results revealed that in decreased PLCE1 expression was associated with tumor progression in HCC and may function as a promising biomarker for HCC prognosis.
