Breast cancer in the malignant tumor,not only high incidence,mortality has been high,and breast cancer has always been a threat to the global primary disease in women.Female breast is composed of skin,composed of fibrous tissue,mammary gland and fat,breast cancer occur in mammary gland malignant tumor of epithelial tissue.Breast cancer,99% in women,only 1% in men.Mammary gland is not to maintain life activities of the important organs,in situ breast cancer is not mortal;But cause to loss of breast cancer cell characteristics of normal cells,connection between the cells and cells become loose,so fall off easily.Cancer cell falls off at once,free with the lymph or blood,spread of cancer cells through off the body,form a transfer,life-threatening factor.The current common tumor of breast cancer has become a threat to women's physical and mental health.Chronic stress was involved in the metabolic process in many diseases,endoplasmic reticulum is one of the widely exists in eukaryotic cells important organelles,is mainly responsible for protein synthesis and transport,folding,assembly,under the stimulation of external factors,cells under stress,cause not fold proteins reaction,it is a kind of self protection mechanisms,the collective but when stress continues,will induce inflammation or reaction,such as apoptosis,and triggered a series of pathological reaction.Endoplasmic reticulum Stress(ERS)occurs at a earliestcellular level.Recent studies have shown that endoplasmic reticulum stress is associated with many diseases in humans,such as neurodegenerative diseases,metabolic diseases,the pathogenesis of inflammation and cancer.Recent study found that endoplasmic reticulum research is closely related to the malignant progression of breast cancer,but the specific influence mechanism is not clear.So further study of the pathogenesis of breast cancer,looking for the ideal for clinical treatment of breast cancer drugs,has important theoretical and clinical significance.Endoplasmic reticulum stress model induced by low-dosage tunicamycin is one of the classic model.This topic first focused on the endoplasmic reticulum stress,the changes of the changes of endoplasmic reticulum stress related protein,then test the biological behavior of breast cancer cells.We found that low-dosage TM induced group,the expression of GRP78,p-eIF2 a,XBP-1S,which related to endoplasmic reticulum stress,is enhanced.And then tested the malignant biological behavior of them,we found that low-dosage TM induced group,cell proliferation,migration,malignant biological behavior is enhanced.And has a dose dependent.The malignant degree of50 nM induced group is significantly higher than 10 nM.We suspect that the malignant biological enhancement may be due to the endoplasmic reticulum stress in breast cancer cells.In conclusion,this study result shows that breast cancer cell lines MCF-7 and MB-231 after low-dosage TM treatment,ERS was activated,and then proliferation,migration,invasion and other malignant biological behavior ability have been enhanced,ultimately promote the invasive breast cancer metastasis.This may due to low-dosage TM can induce endoplasmic reticulum stress in breast cancer cells,endoplasmic reticulum stress of breast cancer and its specific mechanism remains to be explored.As we know,4-PBA can block the endoplasmic reticulum stress.So we plan to use4-PBA pre-treat MCF-7 and MB-231 endoplasmic reticulum stress model,then testmalignant biological behavior of MCF-7 and MB-231,to verify that the enhancement of malignant biological behavior of breast cancer is due to endoplasmic reticulum stress.
