Expression And Role Of SALL2 Gene In Oral Tumor

Objective Sal-like gene 2(SALL2)is a tumor suppressor gene.It can inhibit the proliferation of tumor cells,promote tumor cell apoptosis.To detect the expression levels of SALL2 in oral tumor,and to explore its influence on the proliferation and migration of oral tumor cells.To investigate whether SALL2 can be used as a tumor marker and play a role in the treatment of oral tumor.Methods The study constructed SALL2 gene overexpression and knockdown of oral tumor(SCC-3)cell line and observed the changes of cell proliferation and migration by MTT test and cell scratch test.15 groups of oral tumor tissue and tumor adjacent tissue were collected,and the expression of SALL2 m RNA was detected by QRT-PCR method.Results1.The plasmid containing the target gene was designed and synthesized,and the plasmid was introduced into the oral tumor cells by lentivirus packaging.By using this method,we can construct an oral cancer cell line with stable overexpression or knock down.2.The scratch experiment compared the results of SCC-3 cell lines in each group to scratch the intermediate growth speed of movement,can inhibit oral tumor cell migration effect of overexpression of SALL2 gene,while knockdown of SALL2 gene with enhanced oral tumor cell migration.3.MTT assay was used to detect the growth curve of SCC-3 cells in each group.The over expression of SALL2 gene could inhibit the proliferation of oral cancer cells,while knockdown of SALL2 gene could enhance the proliferation of oral cancer cells.4.QRT-PCR method was used to detect the expression level of SALL2 m RNA in 15 groups of specimens.The results showed that the expression level of SALL2 in oral tumor tissue was decreased compared with that in normal tissues.Conclusions1.SALL2 is involved in the occurrence and development of oral cancer,through a certain mechanism of oral tumor cell cycle regulation,inhibition of tumor cell migration and growth.2.SALL2 can be used as a potential tumor marker for oral cancer to judge the biological behavior of tumor cells,to monitor tumor metastasis,to evaluate the therapeutic effect and prognosis.3.The expression of SALL2 in oral tumors regulating mechanism,biological function and specific mechanism of inhibition of tumor metastasis has not been elucidated.This study provides a theoretical basis for the further study of the mechanism of SALL2 in oral tumor.
