Objective To study the expression level of PJA1 and mi R-130 a in oral tumor tissues and to determine the effect on tumor cell migration and proliferation,and to analyze the possible roles of the two genes in the development and progression of tumor,and to explore the interaction between the two genes.Methods Collected 15 groups of oral tumor and adjacent tissues,and extracted their m RNA.The expression level of PJA1 and mi R-130 a was detected by QRT-PCR.Overexpression of PJA1,overexpression of mi R-130 a,overexpression of both PJA1 and mi R-130 a in SCC-3 were constructed.The effect of cell proliferation was examined by MTT assay,and the effect of cell migration was determined through Wound scratch assay.Results QRT-PCR revaled that the expression of PJA1 in oral tumor tissues was obviously lower than that in normal tissues(P <0.05),the expression of mi R130 a in oral tumor tissues was significantly higher than that in normal tissues(P <0.05).MTT and Wound scratch assay demonstrated that overexpression of PJA1 could inhibit the migration and proliferation of SCC-3,overexpression of mi R130 a could promote the migration and proliferation of SCC-3.PJA1 could inhibit the ability of mi R130 a to promote the migration and proliferation of SCC-3 when both of them were expressed at the same time.Conclusion The expression level of PJA1 was down-regulated in oral tumor tissues,PJA1 could inhibit tumor cell proliferation and migration.The expression level of mi R130 a was up-regulated in oral tumor tissues,mi R130 a could promote tumor cell proliferation and migration.mi R130 a might promote tumor cell proliferation and migration in part by inhibiting PJA1.
