| Objective:Access and collate literature related to depression and insomnia,identify diseases similar to depression and insomnia,and classify these diseases as depression and insomnia.According to the clinical symptoms,the etiology and pathogenesis of insomnia,the relationship between the syndrome and the insomnia,and discuss the theoretical basis of Qi depression.To observe the effect of Sanshao Anshen Decoction on insomnia and its influence on neurotransmitter and related neurotransmitter receptors in depressive insomnia animal model.The theoretical basis and experimental basis of the clinical application of San cao An shen Prescription were provided through theoretical discussion and experimental demonstration.Method:1.Theoretical discussion:review and organize the literature review system and review the insomnia related literature depression,insomnia and depression to find similar diseases,and the sorting and classification of such diseases.From the theory of Chinese medicine to clear the relationship between depression and insomnia,Qi from the theory of depression on the theory of insomnia.2.Experimental study:Forty SD male Sprague-Dawley rats were used to establish the animal model of depressive insomnia by chronic unpredictable moderate stressmethod,solitary-culture method and multi-platform water environment sleep deprivation method.The body weight and behavioral changes were observed.Morris water maze was used to test the ability of learning and memory in rats.The rat neurons of the blank group,the model group,the western medicine group and the Sanqi Anshen group were compared by HE staining.The levels of monoamine neurotransmitters 5-HT and NE in serum and cerebral cortex of rats were measured by enzyme-linked immunosorbent assay(ELISA).Real time-PCR was used to detect the expression of serotonin receptor,γ-aminobutyric acid receptor and dopamine receptors in hypothalamus and hippocampus.Results:1.Body mass test results:During the modeling of normal weight group rats with time changed and increased slowly,without depression insomnia when modeling(T=0)made of body mass model rats and the normal control group rats compared with body weight had no difference(P>0.05),model(T>0)made the weight module the rat is not changed when modeling and have increased body weight of rats,then made module gradually stabilized,from the 7D to build weight model rats and the normal control group at the same time the rats compared with the differences(P<0.01).The 28 D and the 35 D when the weight of rats in model group were significantly lower than the normal group(P<0.01),western medicine group and the three group in the grass and to give the corresponding drug,the body weight of rats increased significantly,and compared with the model group,the differences were statistically significant(P<0.01,P<0.05).Sucrose preference experiment results : There was no significant difference in the sucrose preference between the untreated group(T = 0D)and the model group(P> 0.05).The sucrose preference in the 7D model group was lower than that in the normal group(P <0.05).The sucrose preference of therats in the model group was significantly lower than that of the normal rats(P<0.01).The preference of sucrose in the western medicine group and Sanshao Anshenfang group was improved(P <0.01,P <0.05)compared with the model group after administration of 7D(28D)and 14D(35D)Sucrose preference was significantly lower than that of normal rats(P <0.01).2.Morris water maze test results:Compared with the model group,the rats of western medicine group and San Cao An Shen Prescription group to find the platform time and swimming distance decreased(P<0.01,P<0.05),western medicine group and San Cao An Shen Prescription group rats the number of cross platform and target quadrant time increased(P<0.01,P<0.05),western medicine group and San Cao An Shen Prescription group rats in group first times against the original platform also shortened(P<0.01,P<0.05).Results of autonomous activity experiment:Compared with the blank group,the number of activities and the distance of movement of the model group were significantly lower than those of the blank group(P <0.01).Compared with the model group,the number of activities and the distance of movement of the western medicine group and San Cao An Shen Prescription group were all improved(P <0.01,P <0.05).3.HE staining results: Nerve cells in the nucleus deep staining,blank group of neuronal cells arranged in the rules,even more uniform distribution Compared with the blank group,the neurons in the model group were irregular or disordered,the number of neurons was decreased and the nucleus was reduced.Compared with the model group,the neurons of the blank group were more Compared with the western medicine group and San Cao An Shen Prescription group of neuronal cells were significantly increased and arranged more rules.4.ELISA test results:Compared with the blank group,the content of5-HT in the serum and cerebral cortex of the model group decreasedsignificantly(P <0.01),NE content in serum and cerebral cortex No significant change(P> 0.05).Compared with the model group,the contents of5-HT in the serum and cerebral cortex of the western medicine group and San Cao An Shen Prescription group were significantly increased(P<0.01).Compared with the blank group,the contents of Glu and GABA in the serum and cerebral cortex of the model group were significantly increased(P<0.01),and the Glu/GABA ratio also increased(P<0.01).Compared with the model group,the contents of Glu and GABA in the serum and cerebral cortex of the western medicine group and San Cao An Shen Prescription group decreased(P<0.01,P <0.05),and the Glu/GABA ratio also decreased(P<0.01).5.Real Time-PCR detection results:Compared with the blank group,the expression of 5-HT1 AR mRNA in the hypothalamus and hippocampus of model group decreased significantly(P<0.01),while the expression of5-HT2 AR mRNA in hypothalamus and hippocampus increased significantly(P<0.01).Compared with model group,the expression of 5-HT1 AR mRNA in hypothalamus and hippocampus were increased(P<0.01,P<0.05),and the expressions of 5-HT2 ARmRNA in hypothalamus and hippocampus were significantly increased(P<0.01,P <0.05).Compared with the blank group,the expression of GABAAR_α1 and GABAAR_γ2 mRNA in the hypothalamus and hippocampus of the model group were significantly increased(P<0.01).Compared with model group,the expression of GABAAR_α1 mRNA and GABAAR_γ2 mRNA in hypothalamus and hippocampus of western medicine group and San Cao An Shen Prescription group were decreased(P<0.01,P<0.05).Conclusion1.The theory of TCM contains in-depth understanding of circadian rhythm based on the opinion such as culturecorrespondence between man anduniverse,change of Yin-yang,supersedure of Wuxing,conversion of Yingwei.The theory of circadian rhythm is widely used indiagnose,treetment and prognosis in TCM.2.Chronic unpredictable moderate stress method combined with solitary method and combined with multi-platform water environment sleep deprivation depression model of insomnia animal model is replicable3.San Cao An Shen Prescription has antidepressant and sedative and hypnotic effects,can improve the depression and sleep status of the insomnia rats;At the same time,San Cao An Shen Prescription can improve the learning and memory ability of the insomnia rats.4.The depression,insomnia and learning and memory of insomnia rats may be related to regulating the expression of neurotransmitters and their receptors in the brain. |
