| Objective: This dissertation elaborated the chemical constituents of one Dendrobium plant(Orchidaceae),D.wardianum Warner;Determination of erianinin in twelve species of Dendrobium and investigate the feasibility of erianin in Dendrobium quality evaluation.Methods: 1.The chemical constituents were separated by various column chromatographies(normal phase silica gel,RP-8,Sephadex LH-20,MCI and MPLC,semipro-HPLC).Their structures were elucidated on the basis of extensive spectroscopic methods(1D-and 2D-NMR,MS,IR,UV).2.The content of erianinin in twelve species of Dendrobium was determined by LC-MS/MS.Results: 1.23 compounds were isolated and identified from the stems of D.war dianum Warner,including aduncin(1),wardianumine A(2),dihydroshihunine(3),denbinobin(4),9,10-dihydro-5-hydroxy-3,7-dimethoxy-1,4-phenanthrenequinone(5),mostatin(6),loddigesiinols A(7),moscatilin(8),5-hydroxy-3,4′-dimethoxybiben zyl(9),3,4-dihydroxy-5,4’-dimethoxy bibenzyl(10),dendrocandin A(11),gigant ol(12),dihydroconiferyl dihydro-p-coumarate(13),9-[7-[(7′S,8′S)-7′-(2′-hydroxy-1′-dimethoxyphenyl)-8′-hydroxymethyl-1-methoxy-7′,8-dihydrobenzo[2a,3a]dioxin-5-yl]ethyl]-12-methoxylbenzene(14),dendrocandin U(15),ethyl 3-(4-hydroxyphenyl)acrylate(16),pinoresinol(17),(+)-medioresinol(18),episyringaresinol(19),(+)-Syringaresinol(20),3′,4′,5′-trimethoxyflavanone(21),homoeriodictyol(22),protoc atechuic acid methyl ester(23).The types of these structures include sesquiterp enes,bibenzyls,alkaloids,phenanthrenes,flavones and lignanoids;Compounds 1,2,3,8 were evaluated for the inhibitory activieties on nitric oxide production.The results showed that the four compounds had little activity.Cytotoxicity tests were performed on compounds 4,5,6,14,the results showed that compound 5 have better cyctltlxicity against MCF-7 and SW480,compound 14 have good se lectivity for MCF-7.2.Erianin were only detected in three species of Dendrobium(Dendrobium chrysotoxum,Dendrobium nobile and Dendrobium fimbriatum),erianin in the given concentration ranges present a good linear relationship(r=0.9994),and the contents of three species of Dendrobium were 0.159%,0.0564%,0.0392%.Conclusion: 1.23 compounds,including 2 new ones(2 and 14),were isolated from D.wardianum Warner.2.The method is sensitive,reliable,and rapidly for determination of erianinin in Dendrobium,which providing a scientific basis for further research and development and quality control standard of Dendrobium.In addition,the progress on chemical constituents of the sesquiterpenes from Dendrobium plants was reviewed,these compounds are summarized based on these structural features,which providing a theoretical basis for further research and development of Dendrobium. |
