1 Clinical research1.1 Objective: To study the correlation serum of ROCK1 in COPD patients complicated with pulmonary arterial hypertension.1.2 Methods: Collected the clinical data of COPD patients who were hospitalized in Anhui University of Traditional Chinese Medicine First Affiliated Hospital from February 2016 to December 2016.All patients accepted pulmonary arterial systolic pressure test by echocardiography.According to the Guideline of pulmonary hypertensionby ESC in 2009 and Exclusion criteria,15 COPD patients without pulmonary arterial hypertension were taken as group B,And 15 COPD patients who were complicated with pulmonary arterial hypertension were selected as group C.15 people who take health examination were chosen to group A.The levels of ROCK1 in serum of all objects were tested by enzymelinked immunosorbent assay(ELISA).The correlation between ROCK1 and PASP in patients with COPD was analyzed.1.3 Results The pulmonary arterial systolic pressure(PASP)in the three groups were25.43±6.12 mm Hg,27.13±6.48 mm Hg and 58.10±9.31 mm Hg.Serum ROCK1 expression levels in the group A?B?C were 22.3±6.65 pmol/L,24.1±5.28 pmol/L and 36.7±6.11pmol/L.The levels of ROCK1 in serum of group C were higher than those of group A and group B(P<0.05).There was no difference between group B and group A(P>0.05).There was a positive correlation between the serum ROCK1 and the pulmonary arterial systolic pressure(r=0.637,P<0.05).1.4 Conclusions: The expression of serum ROCK1 increases in COPD patients with pulmonary arterial hypertension.It may be a marker of the severity of patients with PAH realated COPD.2 Experimental study2.1 Objective By investigating the effects of Shen Qihua Tan on Bcl-2 and Bax Protein expression in COPD model rats,and the intervention of Rho/Rho kinase signal pathway,to explore the molecular mechanisms of prevention and treatment airway remolding with Shen Qihua Tan.The purpose is to provide a scientific theory basis for clinical treatment.2.2 Methods SD rats were randomized into healthy rats and COPD rats model by spss software.The COPD rats model was performed with intratracheal instillation of lipopolysaccharide(LPS)on the first and 14 th day and exposed to cigarette smoke for60 days,30 minutes per day.Pathology to determine the model successfully reproduced,the COPD rats model were randomized into normal,model,jinshuibao,Shen Qihua Tan high?middle and low dose groups.normal,model groups were given the same amount of distilled water every day.Jinshuibao group was given adult equivalent dose of drug.Shen Qihua Tan high?medium and low dose groups of were respectively given each drug according to the adult dose of 14 ?7?3.5 times,for four weeks.Then observed every group rat lung function ? lung tissue pathological by HE staining.Immunohistochemical method was developed for the determination of the Bcl-2 and Bax in lung tissue.RT-q PCR was developed for the determination of the Rho A m RNA?Rock?/?m RNA in airway smooth muscle tissue.2.3 Results2.3.1 Compared with the normal group,the rest of the groups Bax expression in rat lung tissue were significantly reduced.There are significantly different between these groups(P<0.05).Compared with model group,the treatment groups Bax expression in rat lung tissue have increased,the difference was statistically significant(P<0.05).Compared with normal group,the rest of the groups Bcl-2 were significantly increased in the lung tissue(P<0.05).Compared with model group,the treatment groups Bcl-2 were reduced in the lung tissue,there are significantly different between these groups(P<0.05).Compared with jinshuibao group,Shen Qihua Tan high?middle and low dose groups Bax have increased in the lung tissue,the difference was statistically significant(P<0.05).Compared with jinshuibao group,Shen Qihua Tan high?middle and low dose groups Bcl-2 is reduced in the lung tissue,the difference was statistically significant(P<0.05)2.3.2 Compared with the normal group,the rest of the groups Rho A m RNA,Rock?/?m RNA were significantly increased in lung tissue(P<0.05).Compared with model group,the treatment groups Rho A m RNA,Rock?/? m RNA were lower in the lung tissue,the difference was statistically significant(P< 0.05)and High doses reduce the most obvious.Compared with jinshuibao group,Shen Qihua Tan high?middle and low dose groups Rho A m RNA,Rock?/?m RNA were reduced in the lung tissue,there are statistically significant between these groups(P<0.05)2.4 Conclusions ShenQihuaTan can through the RhoA/Rho signal pathway,can cut the Rho A m RNA,Rock?/?m RNA expression in rat lung tissue,reduce the Bcl-2expression,increase the expression of Bax,improve lung function,slow down the formation of COPD airway remodeling.
