Research On Immunologic Mechanism Of Invigorating Qi For Ascending Method On Rats With Experimental Autoimmune Myasthenia Gravis

Objective:To investigate the immune mechanism of Invigorating Qi for ascending method in EAMG rats.Methods:To establish the Experimental Autoimmune Myasthenia Gravis model with Ra97-116.Select 6 rats randomly from 50 SPF female Lewis rats as CFA control group,the remaining 44 rats were immunized,36 of which were successful after assessment.They were randomly divided into 6 groups:EAMG model group,Positive control(prednisone),Shengxian decoction group,Invigorating Qi for ascending group(Astragalus membranaceus,Bupleurum chinensis,Cimicifugae foetidae,Platycodon grandiflorum),Tonifying Qi group(Astragalus membranaceus),Ascending group(Bupleurum chinensis,Cimicifugae foetidae,Platycodon grandiflorum).Treatment lasted for one month.Rats were weighed every week during the experiment.Clinical symptoms,weight and the decrement percentage of RNS were evaluated.ELISA was adopted to determine the titers of AchR-Ab,IFN-?,TGF-?,IL-17 in serum.FCM was adopted to determine the proportion of CD4+CD25+Foxp3+Treg in peripheral blood lymphocytes of rats.Results:After modeling,compared with the CFA control,body weight of each group rats went down notably,and the symptoms of myasthenia were obvious.The levels of AChR-Ab going up evidently(P<0.01),the percentage of decrement of RNS of each group rats noticeably increased by more than 10%in comparison with that in CFA control(P<0.01),it proves that the EAMG model succeeded.After medication,compared with the model group,in Shengxian decoction,Invigorating Qi for ascending,Positive control groups,the percentage of decrement of RNS of rats decreased significantly(P<0.01 or P<0.05),body eight and other symptoms were improved.The levels of AChR-Ab,IFN-? and IL-17 descended evidently,with TGF-? level going up evidently(P<0.01 or P<0.05);the proportion of CD4+CD25+Foxp3+Treg in peripheral blood lymphocytes increased notably(P<0.05).Conclusion:Invigorating Qi for ascending method can bring an improvement to body symptoms in rats of EAMG.Also,it can increase the proportion of CD4+CD25+Foxp3+Treg and TGF-? level,decrease the levels of IFN-? and IL-17,so as to decrease AChR-Ab level in the blood serum.The immune mechanism of it may be restraining the synthesis or promote the decomposes of AChR-Ab,by regulating the proportion of lymphocyte to enhance immune suppression and reduce autoimmune response.
