Objective To observe the effect of grilled nux vomica on the level of AchRab,IL-6 and TGF-β1 of rat experimental autoimmune myasthenia gravis(EAMG),and explore the therapeutic effect and preliminary mechanism of grilled nux vomica on patients with myasthenia gravis(MG).Methods After takeing 8 rats from 50 female lewis rats randomly as normal group,the EAMG animal models were established in the rest 42 rats immunized by the rat sequence 97-116 of the AChR a subunit(R97-116),40 of which were successful. According to the clinical scored,they were randomly divided into 5 groups.EAMG model group, positive group, grilled nux vomica low、medium、high dose group. The low,medium and high dose of grilled nux vomica were 75mg/(kg-d),150mg/(kg-d),225mg/(kg-d),and the positive group was given prednisone 4mg/(kg-d),once daily.The normal group and model group were administered to the distilled water.4 weeks later,the weight and the clinical symptoms were evaluated,ELISA were adopted to determine the titers of AchRab, IL-6 and TGF-β1 in serum.Results (1)Except the normal group, most of the rats had different degrees of muscle weakness after R97-116 immunized, with the time dragged on the more symptoms they were.There were two rats died during the model building, One died of seriously muscle weakness symptoms,another one was killed by others because of bleeding under the skin. According to Lennon symptom scores,there were 26 rats scored 1.51evel and 9 rats scored 21evel,the positive rate was 85.71%.Neostigmine test results were all positive,the positive rate was 97.62%. So it was show that the EAMG model were built successful,and the muscle weakness scores were significantly different from the normal rats.(2) The levels of AchRab and IL-6,TGF-β1 in the model group were significantly higher than the normal group(P< 0.01). After the pharmacological intervention,the level of each group was decreased.The level of AChRab in high dose group was remarkablely decreased than the medium and the low dose group of grilled nux vomica(P<0.01), but it had no difference to prednisone group(P>0.05). The level of IL-6 in each group of nux vomica was significantly lower than prednisone group,especially the high and the middle dosage groups(P<0.01). The level of TGF-β1 in the medium and the high dose group of grilled nux vomica were significantly lower than the prednisone group(P<0.01). The higher the drug dose of the grilled nux vomica, the lower the level of TGF-β1 in these grilled nux vomica groups(P<0.01orP<0.05). (3) The rats’weight were significantly increased in the model group,the medium and the low dose of grilled nux vomica groups after treatment as compared with that before treatment. (P<0.01orP0.05), the medium dose group of grilled nux vomica were significantly higher than that in the prednisone group(P<0.05).Conclusion(1)Based on the comparability of symptoms scores,the neostigmine test results,the levels of AchRab and IL-6,TGF-β1 between normal group and the others, AChR-a97-116 can serve as the antigen to induce EAMG in rats. (2)It is suggested that grilled nux vomica has effect on anti EAMG. The mechanism is to reduce the levels of AchRab and IL-6,so as to reduce the damage to the postsynaptic membrane AchR, which can alleviate the EAMG condition. (3)The grilled nux vomica has effect on EAMG,the mechanism might be regularize the level of TGF-β1 to maintain the dynamic balance of the body’s immune activation and immunosuppression. Dose within the scope of the grilled nux vomica is safe and effective, especially in improving the weight of EAMG body better, regulating immune function, etc.
