Objective:Study the effects of acupuncture on pain behavior and mechanism of acupuncture on improving chronic inflammation.Methods:We randomly divided 24 C57BL/6J male mice into three groups.The model and acupuncture groups were administered with lipopolysaccharide(LPS)3mg.kg-1 by intraperitoneal injection to induce chronic inflammation,while the control was administered with saline 4ml.kg-1 by intraperitoneal injection.Acupuncture group were treated by acupuncture at bilateral zusanli points(ST 36).After the different treatments of three groups,the Von Frey fiber filament was used to detect the mechanical withdrawal threshold(MWT)of the mice in each group.The content of tumor necrosis factor-a(TNF-a)in each group was detected by ELISA.Bilateral dorsal root ganglion(DRG)neurons were taken to detect the transient receptor potential vanilloid 1(TRPV1)protein expression differences by western blotting.The expression of TRPV1 positive cells of mice DRG of each group were detected by Immunohistochemical staining.Results:Compared with model group,acupuncture could increase the MWT significantly(P<0.05),decreased the serum TNF-a content significantly(P<0.05),decreased TRPV1 protein expression of DRG neurons(P?0.05)and decreased the expression of TRPV1 positive cells of DRG neurons(P<0.05).Conclusion:Acupuncture treatment could reduce the expression of inflammatory factors,but also changed the expression of in DRG neurons and transient receptor potential vanilloid 1(TRPV1),so as to suppress the pain caused by chronic inflammation.
