Expression Of TREM-1 In Mouse Inflammatory Mode Induced By P.acnes And Study Of Effect Of Loquat Qingfei Drink On Inflammation

Objective: Through testing the expression of TREM-1 in the animal former of mice acne,To investigate the effect of TREM-1 on acne inflammation.Using the loquat Qingfei drink treat acne model,and Skin lesions,tissue pathology,the expression of tissue TREM-1,IL-1?,and TNF-? were measured,And explore the loquat Qingfei drink mechanism for the treatment of acne vulgaris.Methods: sixty mice aged from 6 to 8 weeks,randomly divided into six groups: PBS control group,model group,positive control group,loquat Qingfei drink high dose group,middle dose group and low dose group,with 10 in each.The animal former of mice acne was set up by P.acnes,then treated mice by different drugs,PBS control group and model group gives physiological saline,positive control group gives tetracycline,the dose was 0.129g/kg/d,loquat Qingfei drink high dose group,middle dose group and low dose group gives water decoction of loquat Qingfei drink,the doses were 50.72g/kg/d,25.36g/kg/d,and 12.68g/kg/d.the animals in each group orally administered once a day.each time 1ml,The treatment lasted for 14 days.we observe auricle lesions and thickness during the treatment,The tissue pathology,expression of tissue TREM-1,IL-1? and TNF-? were measured after treatment.Results: Analysis of the results of skin lesions and auricle thickness before and after treatment,The PBS control group and model group had no marked changes,the skin lesions and auricle thickness all lessened in the other groups after treatment.The positive control group,high dose and moderate dose traditional Chinese medicine group had the most significant improvement.the skin lesions and average thickness in the other groups were significantly increased compared with PBS control group before treatment(both P?0.05),Compared with model group,the skin lesions and average thickness were decreased in the other groups after treatment,differences are significant(both p<0.05).Analysis of the results of pathology,There were no abnormal pathological changes in PBS control group,The Pathology of model group showed that these were apparent tissue edema and Vascular congestion,and there were a large number of inflammatory cell infiltration and abscess in the auricular tissue.Compared with model group,the vascular congestion,inflammatory cells and abscess were lessened in the other groups after treatment.Analysis of the results of detecting by Western blot and RT-PCR,the expression of auricle tissue TREM-1 in the model group is significantly increased compared with PBS control group,differences are significant(P?0.05),Compared with model group,the expression of auricle tissue TREM-1 all decreased in the other groups after treatment,especially the positive control group,high dose and moderate dose traditional Chinese medicine group.differences are significant(both p<0.05).Analysis of the results of detecting by ELISA,The serum IL-1? and TNF-? levels in model group were remarkably higher than in the PBS control group,differences are significant(p<0.05),Compared with model group,the levels of inflammatory factors all decreased in the other groups after treatment,especially the positive control group,high dose and moderate dose traditional Chinese medicine group.differences are significant(both p<0.05).Conclusion: 1.TREM-1 take part in the inflammation immunologic reaction of acne model,and correlated to the seri ousness of the acne model inflammation.2.TREM-1 may regulate the conditions of inflammatory reaction by affect acne inflammatory signal transduction,IL-1? and TNF-?.3.loquat Qingfei drink has a marked suppressive effects on the inflammation of acne model.high dose,moderate dose and low dose traditional Chinese medicine group were to make the Acne Model TREM-1,IL-1? and TNF-? levels decreased.4.loquat Qingfei drink may decrease the level of TREM-1,IL-1? and TNF-? which had anti inflammation effects.
