Objective:To investigate the expression and relationship between pleiotrophin(PTN)and gastric cancer tissue and peripheral blood in patients with gastric cancer.The role of PTN in the occurance of gastric cancer and its role in gastric cancer diagnosis,treatment strategy and prognosis.Methods:1.Collecting cancer tissues(group A)and para-cancer tissues(group B)of 70 cases with stomach carcinoma who underwent surgery during Mar.2015 to Dec.2016.These cases were chosen as the experimental group(n=70),and collected30 normal persons as the control group(n=30).Inclusion criteria:(1)Stomach carcinoma diagnosed by histopathology examination and gastroscopy before operation;(2)Would prepared to be performed operation;(3)Patients without important viscera dysfunction,having tolerance to surgical operation;(4)Volunteered to surgical operation.Exclusion criteria:(1)Patients with stomach carcinoma recurrence,residual gastric cancer or other cancer;(2)With a history of gastro-intestinal surgery,gastric ulcer or enteritis;(3)With vital organs dysfunction sunch as heart failure or pulmonary dysfunction;(4)Patients with diseases of the hematopoietic system;(5)Used immunosuppression drugs hormone or blood products recently;(6)Associated with severe malnutrition;(7)Preoperative chemotherapy,radio therapy,immune therapy or other treatment.2.Obtained the gastric cancer tissues and para-cancer tissues(from tumor tissue>5cm)during operation.These samples were tesed by immunohistochemical technique,and analysed the relationship between PTN expression and clinicopathological features of gastric cancer.3.With enzymelinked immunosorbent assay(ELISA),Collected peripheral blood samples of70 normal persons(the control group)and 30 cases of gastric cancer(gastric cancer group).Analysed the relationship between PTN expression and clinicopathological features of gastric cancer.were selected to test the serum PTN.Results:1.The positive expressions of PTN in in group A(81.4%)is all significantly higher than that of group B(11.4%),with a significant difference(P<0.05).The PTN expression in tumor size≥4cm is higher than that in tumor size<4cm,PTN is highly expressed in patients with TNM stage Ⅲ-Ⅳcompared with TNM stage Ⅰ-Ⅱ(P<0.05),the PTN expression is lower in Borrmann0-Ⅱcompared with BorrmannⅢ-Ⅳ(P<0.05),the expression of PTN is highly closed with lymphatic metastatisis(P<0.05).But with different genders and ages,there were no significant differences of PTN expression(P>0.05).2.The serum PTN were all significantly higher of gastric cancer group than that of the control group(P>0.05).Serum PTN is higher in patients with TNM stageⅢ-Ⅳcompared with TNM stage Ⅰ-Ⅱ(P<0.05),the serum PTN is highly closed with lymphatic metastatisis(P<0.05).Histological differentiation degree,genders and ages don’t have influence on serum PTN expression(p>0.05).Conclusion:1.The level of PTN expression in the gastric cancer tissue was significantly higher than that of the further-tumor tissues,so it can determine the positive and micrometastasis of the surgery.2.The serum PTN were all significantly higher of gastric cancer group than that of the control group.Clinical pathologic features such as tumor size,TNM staging and lymph node metastasis are associated with gastric cancer patients and are expected tobe a test for the diagnosis of gastric cancer.3.The expression of PTN in gastric cancer tissue is related to tumor staging,type,and differentiation,indicating that PTN can be used for diagnosis of gastric cancer,strategy formulation and prognosis.4.PTN provides evidence for the screening and diagnosis of gastric cancer.
